摘要:
DNA是遗传信息的载体,其结构的稳定性对维持生命至关重要。当内源性或外源性因素引起DNA损伤时,细胞会启动DNA损伤反应(DDR)机制进行修复,从而使细胞停留在分裂间期或死亡。但在肿瘤细胞中,往往存在DDR抑制或突变,导致肿瘤细胞恶性增殖。极光激酶(AURK)家族是一类与有丝分裂过程密切相关的蛋白激酶。研究发现,AURK家族在肿瘤细胞中出现异常表达,并且与DDR相关分子p53和细胞周期蛋白依赖性激酶(CDKs)存在密切联系。因此,现就DDR和AURK家族在肿瘤发生与发展中的作用以及两者在肿瘤中的关系进行综述。
Abstract:
DNA is the carrier of genetic information,and the stability of its structure is essential for the maintenance of life.When DNA damage is caused by endogenous or exogenous factors,the cell will initiate the DNA damage response(DDR)mechanism to repair it,thus allowing the cell to stay in interphase or die.However,in tumour cells,DDR inhibition or mutations are often present,leading to malignant proliferation of tumour cells.Aurora kinase(AURK)family is a class of protein kinases that are closely related to the mitotic process.It has been found that that the AURK family is abnormally expressed in cancer cells and has a close relationship with DDR-related molecules such as p53 and cyclin-dependent kinases(CDKs).Therefore,this article reviews the roles of DDR and AURK family in tumourigenesis and development and the relationship between the two in cancer.
