摘要:
目的:探讨三氧化二砷对人肺癌细胞NCI-H460增殖、凋亡的影响及其可能的分子机制。方法:体外培养人肺癌细胞株NCI-H460,利用CCK-8法、集落形成实验、流式细胞技术分别检测三氧化二砷对NCI-H460细胞的增殖、集落形成、细胞周期和凋亡,Western bolt检测相关细胞周期因子和凋亡蛋白的表达。结果:三氧化二砷能够明显抑制NCI-H460细胞的增殖、阻滞细胞周期、诱导细胞凋亡,并且呈浓度依赖性。Western bolt显示:随着药物浓度的增加,B淋巴细胞瘤-2蛋白(Bcl-2)、周期蛋白依赖性激酶-1(CDK1)、原癌基因(c-Myc)表达下调,BCL2相关蛋白-x(Bax)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、Caspase-9表达上调。结论:三氧化二砷能够抑制NCI-H460细胞的增殖、阻滞细胞周期、诱导细胞凋亡,其机制可能通过下调Bcl-2、CDK1、c-Myc蛋白表达,上调Bax蛋白表达,激活Caspase-9、Caspase-3蛋白酶实现的。
