摘要:
目的:探究磷酸甘油酸激酶1(PGK1)、微小RNA138-5p(miR-138-5p)、微小RNA-195( miR-195)在侵袭性垂体腺瘤(IPT)组织中的表达水平及其与临床特征和预后的相关性。方法:收集168例垂体腺瘤患者为研究对象,并将其分为非IPT组(85例)和IPT组(83例)。比较两组瘤体组织中PGK1、miR-138-5p、miR-195表达水平及其与临床特征、侵袭性积分和术后复发情况的相关性。结果:IPT组瘤体组织中PGK1表达水平高于非IPT组,miR-138-5p、miR-195水平低于非IPT组(均P<0.01)。1 cm≤直径<4 cm患者瘤体组织中PGK1表达水平高于直径<1 cm患者,miR-138-5p、miR-195水平低于瘤体直径<1 cm患者,≥4 cm患者瘤体组织PGK1表达水平高于1 cm≤直径<4 cm患者,miR-138-5p、miR-195水平低于1 cm≤直径<4 cm患者(均P<0.05);功能性垂体瘤和Knosp分级Ⅳ级患者PGK1表达水平高于无功能性垂体瘤和Knosp分级Ⅲ级患者,miR-138-5p、miR-195水平低于无功能性垂体瘤和Knosp分级Ⅲ级患者瘤体组织(均P<0.05)。瘤体组织中PGK1水平与IPT患者侵袭性积分呈正相关,与miR-138-5p、miR-195表达水平呈负相关(r=0.833、-0.499、-0.614,均P<0.001);miR-138-5p、miR-195水平与侵袭性积分呈负相关(r=-0.690、-0.319,均P<0.001),miR-138-5p与miR-195水平呈正相关(r=0.508,P<0.001)。PGK1高表达组、miR-138-5p低表达组和miR-195低表达组的复发率均高于PGK1低表达组、miR-138-5p高表达组和miR-195高表达组(均P<0.05)。结论:IPT患者瘤体组织中PGK1表达上调,miR-138-5p、miR-195表达下调,其表达与患者肿瘤大小、病理类型、侵袭情况及术后复发情况密切相关。
Abstract:
Objective:To investigate the expression levels of phosphoglycerate kinase 1 (PGK1),microRNA-138-5p (miR-138-5p),and miR-195 in the tissues of invasive pituitary adenoma (IPT) and their correlations with the clinical characteristics and prognosis of patients.Methods:168 patients with pituitary adenomas were selected as the research subjects and were divided into the non-IPT group (85 cases) and the IPT group (83 cases).The expression levels of PGK1,miR-138-5p,and miR-195 in the tumor tissues of the two groups were compared.The correlations between the expression levels of each indicator and clinical characteristics,invasiveness score,and postoperative recurrence were analyzed.Results:The expression level of PGK1 in the tumor tissues of patients in the IPT group was higher than that in the non-IPT group,while the expression levels of miR-138-5p and miR-195 were lower than those in the non-IPT group (all P<0.01).The expression level of PGK1 in the tumor tissues of patients with a diameter ranging from 1 to 3.99 cm was higher than that of patients with a diameter less than 1 cm.The expression levels of miR-138-5p and miR-195 were lower in the tumor tissues of patients with a tumor diameter less than 1 cm compared to those of patients with a diameter ranging from 1 to 3.99 cm (all P<0.05).The expression level of PGK1 in the tumor tissues of patients with a diameter of 4 cm or more was higher than that of patients with a diameter ranging from 1 to 3.99 cm,the expression levels of miR-138-5p and miR-195 were lower in the tumor tissues of patients with a diameter of 4 cm or more compared to those of patients with a diameter ranging from 1 to 3.99 cm (all P<0.05).The expression levels of PGK1 in the tumor tissues of patients with functional pituitary adenomas and Knosp grade Ⅳ were higher than those of patients without functional pituitary adenomas and Knosp grade Ⅲ,while the expression levels of miR-138-5p and miR-195 were lower in the tumor tissues of patients with functional pituitary adenomas and Knosp grade Ⅳ compared to those of patients without functional pituitary adenomas and Knosp grade Ⅲ (all P<0.05).The level of PGK1 in tumor tissues was positively correlated with the invasive score of IPT patients,and negatively correlated with the expression levels of miR-138-5p and miR-195 (r=0.833,-0.499,-0.614,all P<0.001);the levels of miR-138-5p and miR-195 were negatively correlated with the invasive score (r=-0.690,-0.319,all P<0.001),and the level of miR-138-5p was positively correlated with the level of miR-195 (r=0.508,P<0.001).The recurrence rates of the high-expression group of PGK1,the low-expression group of miR-138-5p,and the low-expression group of miR-195 were all higher than those of the low-expression group of PGK1,the high-expression group of miR-138-5p,and the high-expression group of miR-195 (Logrank χ2= 6.335,20.839,12.838,all P<0.05).Conclusion:In the tumor tissues of IPT patients,the expression of PGK1 is upregulated,while the expressions of miR-138-5p and miR-195 are downregulated.Their expressions are closely related to the size of the tumor,pathological type,invasion status,and postoperative recurrence of the patients.
