摘要:
目的:探讨鸢尾素(Irisin)对白细胞介素-1β(IL-1β)诱导大鼠甲状腺细胞(FRTL-5细胞)凋亡的影响及可能的作用机制。方法:将FRTL-5细胞随机分为对照组、IL-1β组、IL-1β+Irisin组、IL-1β+Irisin+BAY11-7028[核因子-κB(NF-κB)信号通路抑制剂]组、IL-1β+BAY11-7028组;通过qPCR测定对照组与IL-1β组细胞中Irisin mRNA的表达水平,采用CCK8法检测各组细胞的增殖活性,Western blot检测各组细胞中半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)、BCL2相关X蛋白(Bax)、B细胞淋巴瘤-2蛋白(Bcl-2)、p65和磷酸化p65(p-p65)的蛋白表达水平。结果:IL-1β刺激FRTL-5细胞后,细胞增殖活性下降,Caspase-3、Bax水平升高,Bcl-2、Bcl-2/Bax表达下降,细胞中Irisin mRNA水平降低(均P<0.05)。Irisin可提高IL-1β刺激下FRTL-5细胞增殖活性,下调Caspase-3、Bax水平,促进Bcl-2、Bcl-2/Bax表达(均P<0.05)。Irisin可改善IL-1β刺激下p65的磷酸化,特异性抑制NF-κB(加入BAY11-7082)后,细胞中Bcl-2、Bcl-2/Bax表达下降,Caspase-3、Bax表达上升(均P<0.05)。结论:Irisin能够改善IL-1β抑制的甲状腺细胞增殖活性,并且缓解IL-1β诱导的甲状腺细胞凋亡,其作用机制可能与Irisin对NF-κB信号通路的调节有关。
Abstract:
Objective:To explore the effect and mechanism of Irisin on the apoptosis of rat thyroid cells (FRTL-5 cells) induced by interleukin-1β (IL-1β).Methods:We established control group,IL-1β group,IL-1β + Irisin group,IL-1β+ Irisin+BAY11-7028 group,IL-1β+BAY11-7028 group.FRTL-5 cells were cultured in vitro.The Irisin mRNA in the cells of the control group and the IL-1β group was detected using qPCR.The cell viability was detected with CCK8 method.The expressions of Cleaved Caspase-3 (Caspase-3),BCL2 associated X protein (Bax),B-cell lymphoma 2 (Bcl-2),p65 and p-p65 proteins in cells were detected with Western blotting.Results:After IL-1β stimulated FRTL-5 cells,the cell viability was decreased,the protein levels of Caspase-3 and Bax were increased,the expression levels of Bcl-2 and the ratio of Bcl-2/Bax were decreased,and the mRNA expression of Irisin was also decreased (all P<0.05).Irisin can enhance the cell viability of FRTL-5 cells stimulated by IL-1β,down-regulate the levels of Caspase-3 and Bax,and promote the expressions of Bcl-2 and Bcl-2/Bax (allP<0.05).Irisin declined the IL-1β stimulated phosphorylations of p65.After treatment with the BAY11-7082,the expressions of Bcl-2 and Bcl-2/Bax in cells decreased,while the expressions of Caspase-3 and Bax increased (all P<0.05).Conclusion:Irisin can enhance the proliferation activity of thyroid cells inhibited by IL-1β,and alleviate the apoptosis of thyroid cells induced by IL-1β.The mechanism of its action may be related to the regulation of NF-κB signaling pathway by Irisin.
