摘要:
目的:探讨Ras同源基因家族成员A(RhoA)/Rho相关蛋白激酶(ROCK)信号转导通路在原发性膝关节骨性关节炎(KOA)滑膜组织中的表达及意义。方法:收集30例行全膝关节置换术(TKA)的原发性KOA患者的滑膜组织作为观察组,以25例来自下肢离断术的滑膜组织作为对照组,比较两组样本的外观形态与HE染色病理改变。通过免疫组织化学(IHC)染色、蛋白质免疫印迹(Western blot)及实时荧光定量PCR(RT-qPCR)检测RhoA和ROCK蛋白及mRNA的表达变化。结果:观察组标本呈现高度增生、充血、水肿、细胞排列紊乱及空泡化细胞。对照组标本呈白色,表面光滑无水肿,细胞/基质排列有序且均匀。观察组滑膜炎病理评分及RhoA、ROCK mRNA和蛋白表达水平高于对照组(均P<0.05)。结论:KOA患者滑膜组织中RhoA和ROCK mRNA及蛋白表达水平显著提高,表明RhoA/ROCK信号通路参与了原发性KOA发病过程。
Abstract:
Objective:To investigate the expression and significance of the Ras homolog gene family member A (RhoA)/Rho-associated protein kinase (ROCK) signal transduction pathway in the synovial tissues of primary knee osteoarthritis (KOA).Methods:The synovial tissue samples obtained from 30 patients with primary KOA who underwent TKA and 25 patients who underwent lower limb amputation were set as the observation group and the control group respectively.The appearance morphology and HE staining results of the two groups of specimens were compared.The expression changes of RhoA and ROCK proteins and mRNAs were detected by IHC,Western blot,and RT-qPCR.Results:The observation group specimens showed high proliferation,congestion,oedema,unorganized cellular arrangement,and presence of vacuolated cells.The control group specimens were white with a smooth surface and non-oedematous texture,and showed organized and homogenous cell/stroma arrangement.The pathological score of synovitis in the observation group was higher than that in the control group,as well as the expression levels of RhoA,ROCK mRNA and protein (all P<0.05).Conclusion:Patients with KOA expressed high levels of RhoA and ROCK transcripts and proteins in synovial tissues,indicating that the RhoA/ROCK signaling pathway is involved in the pathogenesis of primary KOA.
