摘要:
目的:探讨余甘子提取物通过蛋白激酶B/哺乳动物雷帕霉素靶蛋白(Akt/mTOR)通路对肺癌细胞A549恶性生物学行为的影响。方法:选取人非小细胞肺癌A549细胞,分为对照组(空白未处理)、余甘子提取物低剂量组(加入50 μg/ml)、中剂量组(加入100 μg/ml)、高剂量组(加入200 μg/ml)、阳性药组(加入100 nmol/L雷帕霉素),采用CCK8法、流式细胞术检测各组A549细胞存活和凋亡率,Transwell小室实验、细胞划痕试验测各组细胞侵袭和迁移能力,Western blot法测各组细胞中Akt、p-Akt、mTOR、p-mTOR蛋白表达。取各组肺癌A549细胞,建立小鼠移植瘤模型,记录各组小鼠移植瘤生长和小鼠存活变化。结果:与对照组比较,低、中、高剂量余甘子提取物组、阳性药组小鼠肿瘤体积、肿瘤质量降低,且呈剂量依赖性(均P<0.05)。低、中、高剂量余甘子提取物组、阳性药组的A549细胞活力、侵袭、迁移能力及p-Akt、p-mTOR蛋白表达均低于对照组(均P<0.05),且阳性药组、高剂量组低于中剂量组,中剂量组低于低剂量组(均P<0.05)。低、中、高剂量余甘子提取物组的A549细胞凋亡率高于对照组,且呈剂量依赖性(均P<0.05)。结论:余甘子提取物可抑制Akt/mTOR通路活性,使肺癌A549细胞侵袭、迁移受到干扰,抑制裸鼠皮下成瘤速度,并诱导细胞凋亡。
Abstract:
Objective:To investigate the effect of Phyllanthus emblica extract on the malignant biological behavior of lung cancer cell A549 through protein kinase B/mammalian target of rapamycin (Akt/mTOR) pathway.Methods:Human non-small cell lung cancer A549 cells were selected and divided into control group (blank untreated),low dose group (50 μg/ml),medium dose group (100 μg/ml) and high dose group (200 μg/ml).CCK8 method and flow cytometry were used to detect the survival and apoptosis rate of A549 cells in each group.Transwell chamber assay and cell scratch test were used to detect the invasion and migration ability of each group.Western blot was used to detect the protein expression of Akt,p-Akt,mTOR and p-mTOR in each group.Lung cancer A549 cells were collected from each group to establish a mouse model of transplanted tumor.The growth of transplanted tumor and survival of mice in each group were recorded.Results:Compared with the control group,the tumor volume and weight of the mice in the low-,medium-,and high-dose Phyllanthus emblica extract groups were decreased in a dose-dependent manner (all P<0.05).The cell viability,invasion and migration abilities and the expressions of p-Akt and p-mtor proteins in A549 cells were significantly lower in the low-,medium- and high-dose EMB groups than in the control group (all P<0.05),and the high dose group was lower than the middle dose group,and the middle dose group was lower than the low dose group (all P<0.05).The apoptosis rate of A549 cells in the low-,medium- and high-dose EmB groups was higher than that in the control group,and showed a dose-dependent manner (all P<0.05).Conclusion:Phyllanthus emblica extract can inhibit the activity of Akt/mTOR pathway,interfere with the invasion and migration of lung cancer A549 cells,inhibit the rate of subcutaneous tumor formation in nude mice,and induce cell apoptosis.
