摘要:
骨关节炎(OA)是一种常见的慢性退行性关节疾病,随着人口老龄化加剧,OA发病率随之上升,严重影响了患者的生活质量,然而其发病机制目前尚未完全明确。软骨细胞凋亡和自噬作为维持软骨细胞内环境稳态的主要机制,在 OA的发生与发展中发挥重要作用。长链非编码RNA(lncRNA)是一类具有超过200个核苷酸的RNA分子,可通过多种机制调控软骨细胞的自噬和凋亡,进而影响OA的进展,如作为竞争性内源RNA(ceRNA)调控微小RNA(miRNA)活性,参与表观遗传修饰、转录调控,以及调节核因子-κB(NF-κB)、磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(AKT)/雷帕霉素靶蛋白(mTOR)等重要信号通路。文章综述了lncRNA在OA软骨细胞凋亡和自噬中作用机制的研究进展,旨在为OA早期的诊断和治疗提供新的见解和靶点。
Abstract:
Osteoarthritis (OA) is a common chronic degenerative joint disease.With the intensification of population aging,the incidence of OA has increased,seriously affecting the quality of life of patients.However,its pathogenesis is not yet fully clear at present.Chondrocyte apoptosis and autophagy,as the main mechanisms for maintaining the homeostasis of the internal environment of chondrocytes,play an important role in the occurrence and development of OA.Long non-coding RNA (lncRNA) is a type of RNA molecule with more than 200 nucleotides.It can regulate autophagy and apoptosis of chondrocytes through multiple mechanisms,thereby affecting the progression of OA,such as regulating the activity of microRNA (miRNA) as competitive endogenous RNA (ceRNA),participating in epigenetic modifications,transcriptional regulation,and regulating important signaling pathways such as nuclear factor-κB (NF-κB),phosphatidylinositol-3-kinase (PI3K)/protein kinase B(AKT)/rapamycin target protein (mTOR).This article reviews the research progress on the mechanism of lncRNA in the apoptosis and autophagy of OA chondrocytes,aiming to provide new insights and targets for the early diagnosis and treatment of OA.
