目的:探讨LINC01123对胰腺癌(PC)细胞增殖、迁移和侵袭的影响及潜在分子机制。方法:收集45例PC患者的PC组织及邻近正常组织样本,体外培养人PC细胞系(PANC-1、SW1990、ASPC1、BxPC-3和CFPAC-1)。实时荧光定量PCR(qRT-PCR)检测组织和细胞LINC01123、微小RNA-185-5p(miR-185-5p)和跨膜蛋白1(TSPAN1)mRNA表达,分析PC组织中LINC01123、miR-185-5p、TSPAN1表达水平的相关性。将PANC-1细胞分为对照组、阴性对照组、LINC01123敲低组、抑制剂对照组和联合组,检测LINC01123、miR-185-5p及TSPAN1 mRNA和蛋白表达,以及细胞活力、细胞凋亡、侵袭和迁移;双荧光素酶报告基因分析PANC-1细胞中LINC01123、miR-185-5p和TSPAN1的调控作用。结果:与正常组织和细胞比较,PC组织和细胞中LINC01123和TSPAN1 mRNA表达升高,miR-185-5p表达降低,且miR-185-5p表达与LINC01123、TSPAN1 mRNA呈负相关,LINC01123表达与TSPAN1 mRNA呈正相关(均P<0.05)。沉默LINC01123可上调miR-185-5p表达,抑制TSPAN1表达,降低PANC-1细胞的增殖、迁移和侵袭能力,并促进细胞凋亡。抑制miR-185-5p可减弱LINC01123沉默对PANC-1细胞恶性生物学行为的抑制作用。双荧光素酶报告基因实验证实,LINC01123可靶向负调控miR-185-5p表达,TSPAN1是miR-185-5p的靶标。结论:沉默LINC01123可能通过miR-185-5p/TSPAN1轴抑制PC细胞恶性生物学行为。
Abstract:
Objective:To investigate the effects of LINC01123 on the proliferation,migration and invasion of pancreatic cancer (PC) cells and its potential molecular mechanism.Methods:PC tissue and adjacent normal tissue samples were collected from 45 PC patients,and human PC cell lines (PANC-1,SW1990,ASPC1,BxPC-3,and CFPAC-1) were cultured in vitro.RT-qPCR was used to detect the expression of LINC01123,miR-185-5p and tetraspanin 1 (TSPAN1) mRNA in tissues and cells.The correlation of LINC01123,miR-185-5p and TSPAN1 expression levels in PC tissues was analyzed.PANC-1 cells were assigned into control group,negative control group,LINC01123 knockdown group,inhibitor control group,and combined group.The expressions of LINC01123,miR-185-5p and TSPAN1 mRNA and protein,as well as cell viability,apoptosis,invasion and migration were detected.Dual luciferase reporter gene experiment was used to analyze the regulatory effects of LINC01123,miR-185-5p and TSPAN1 in PANC-1 cells.Results:Compared with normal tissues and cells,the expression levels of LINC01123 and TSPAN1 mRNA in PC tissues and cells were increased,while the expression level of miR-185-5p was decreased,and the expression of miR-185-5p was negatively correlated with LINC01123 and TSPAN1 mRNA,the expression of LINC01123 was positively correlated with TSPAN1 mRNA (all P<0.05).Silencing LINC01123 was able to upregulate miR-185-5p expression,inhibit TSPAN1 expression,reduce the proliferation,migration and invasion ability of PANC-1 cells,and promote cell apoptosis.Inhibition of miR-185-5p was able to weaken the inhibitory effect of LINC01123 silencing on the malignant biological behavior of PANC-1 cells.The dual luciferase reporter gene experiment confirmed that LINC01123 was able to target negative regulation of miR-185-5p expression,and TSPAN1 was the target of miR-185-5p.Conclusion:Silencing LINC01123 may inhibit the malignant biological behavior of PC cells through the miR-185-5p/TSPAN1 axis.
