摘要:
目的:探究自身免疫性肝炎患者肝组织与外周血T细胞免疫球蛋白及黏蛋白结构域分子3(Tim-3)、细胞因子信号转导抑制因子3(SOCS3)表达及诊断价值。方法:选取自身免疫性肝炎患者83例为自身免疫性肝炎组,选取体检健康者86例为对照组。采用免疫组化染色和酶联免疫吸附法检测肝组织及外周血Tim-3、SOCS3表达。采用全自动生化分析仪检测自身免疫相关的炎症因子白细胞介素-2(IL-2)、IL-6、IL-21以及趋化因子C-C Motif趋化因子配体2(CCL2)、C-X-C Motif趋化因子配体9(CXCL9)、趋化因子C-C-基元受体6(CCR6)表达。采用Pearson相关性分析Tim-3、SOCS3表达与炎症因子和趋化因子的相关性。采用受试者工作特征(ROC)曲线分析Tim-3、SOCS3表达水平对自身免疫性肝炎的诊断价值。结果:免疫组化染色结果显示,自身免疫性肝炎组Tim-3、SOCS3阳性表达率高于对照组(均P<0.05)。与对照组比较,自身免疫性肝炎组炎症因子(IL-2、IL-6、IL-21)、趋化因子(CCL2、CXCL9、CCR6)以及Tim-3和SOCS3水平显著升高(均P<0.05)。Pearson相关性分析显示,Tim-3、SOCS3与炎症因子IL-2、IL-6、IL-21及趋化因子CCL2、CXCL9、CCR6呈正相关(均P<0.05)。ROC曲线分析显示,Tim-3、SOCS3联合检测对自身免疫性肝炎的诊断价值优于各自单独检测(均P<0.05)。结论:Tim-3、SOCS3在自身免疫性肝炎患者肝组织及外周血中均呈高表达,两者联合检测对自身免疫性肝炎具有良好的诊断价值。
Abstract:
Objective:To investigate the expression and clinical significance of T cell immunoglobulin and mucin domain containing molecule 3(Tim-3)and suppressor of cytokine signaling 3(SOCS3)in liver tissue and peripheral blood of patients with autoimmune hepatitis.Methods:Eighty-three patients with autoimmune hepatitis were selected as autoimmune hepatitis group,and 86 healthy people were selected as control group.The expressions of Tim-3 and SOCS3 in peripheral blood and liver tissues were detected by immunohistochemical staining and enzyme-linked immunosorbent assay.Autoimmune-related inflammatory factors IL-2,IL-6,IL-21,CCL2,CXCL9 CCR6 were detected by automatic biochemical analyzer.Pearson correlation analysis was used to analyze the correlation between the expressions of Tim-3,SOCS3 and inflammatory factors and chemokines.ROC curve was used to analyze the diagnostic value of Tim-3 and SOCS3 expression levels for autoimmune hepatitis.Results:Immunohistochemical staining showed that the positive expression rates of Tim-3 and SOCS3 in the autoimmune hepatitis group were higher than those in the control group(both P<0.05).Compared with the control group,the levels of inflammatory factors(IL-2,IL-6,IL-21),chemokines(CCL2,CXCL9,CCR6),Tim-3 and SOCS3 in the autoimmune hepatitis group were significantly increased(all P<0.05).Pearson correlation analysis showed that Tim-3 and SOCS3 were positively correlated with inflammatory factors IL-2,IL-6,IL-21 and chemokines CCL2,CXCL9,CCR6(all P<0.05).ROC curve analysis showed that the diagnostic value of Tim-3 and SOCS3 combined detection for autoimmune hepatitis was better than that of each single detection(all P<0.05).Conclusion:Tim-3 and SOCS3 are highly expressed in liver tissue and peripheral blood of patients with autoimmune hepatitis,and the combined detection of TIM-3 and SOCS3 has a good diagnostic value for autoimmune hepatitis.
