摘要:
目的:探究Ras同源基因家族成员A(RhoA)/Rho相关卷曲螺旋蛋白激酶(ROCK)信号转导通路在膝骨性关节炎软骨组织中的表达及意义。方法:将取自因原发性膝骨性关节炎(KOA)行全膝关节置换术(TKA)去除的30例患者的膝关节软骨组织设为观察组,将同期行下肢离断术去除的25例患者的膝关节软骨组织设为对照组。比较两组标本外观形态,HE染色、番红O-固绿染色、电镜扫描结果。通过免疫组织化学(IHC)染色、免疫印迹(Western blot)及实时荧光定量PCR(RT-qPCR)检测RhoA和ROCK蛋白及mRNA的表达变化。结果:观察组股骨髁软骨表面粗糙,缺损明显,呈溃疡状,染色不均匀,潮线不完整。对照组软骨面光滑平整,组织分层明显,颜色均匀。电镜扫描结果显示,观察组软骨组织毁损严重,结构不清; 对照组表面光洁,软骨间质均匀。与对照组比较,观察组关节软骨组织中RhoA、ROCK蛋白及mRNA表达量高于对照组(均P<0.05)。结论:RhoA/ROCK信号转导通路中的RhoA、ROCK蛋白及mRNA在KOA患者关节软骨组织中表达明显高于行下肢离断术患者,提示RhoA/ROCK信号转导通路与KOA软骨退变具有相关性,可能对KOA的发生与发展起到了重要作用。
Abstract:
Objective:To investigate the expression and significance of Ras homolog gene family member A(RhoA)/Rho-associated protein kinase(ROCK)signaling transduction pathway in cartilage tissue of knee osteoarthritis(KOA).Methods:The knee cartilage tissues of 30 patients with primary KOA removed by TKA were used as the observation group,and those of 25 patients with lower limb amputation removed at the same time were used as the control group.The two groups of specimens were compared in appearance,HE staining,saffranine O-solid green staining and electron microscope scanning.The expressions of RhoA and ROCK protein and mRNA were detected by immunohistochemical(IHC)staining,Western blot and RT-qPCR.Results:In the observation group,the surface of the femoral condyle cartilage was rough,the defect was obvious,ulcerated,the staining was uneven,and the tidal line was incomplete; in the control group,the cartilage surface was smooth and flat,the tissue stratification was obvious,and the color was uniform.The results of electron microscope scanning showed that the cartilage tissue in the observation group was severely damaged and the structure was unclear,and the surface of the control group was smooth and the cartilage mesenchyme was uniform.Compared with the control group,the expression levels of RhoA,ROCK protein and mRNA in the articular cartilage tissue of the observation group were higher than those of the control group(all P<0.05).Conclusion:The expression of RhoA,ROCK protein and mRNA in the RhoA/ROCK signal transduction pathway in the articular cartilage tissue of KOA patients was significantly higher than that in patients undergoing lower limb amputation,suggesting that the RhoA/ROCK signal transduction pathway was associated with KOA cartilage degeneration,which may be related to the occurrence and development of KOA.
