摘要:
目的:探讨子痫前期孕妇微小RNA-210-5p(miR-210-5p)和整合素αM(ITGAM)的表达水平及检测意义。方法:选取40例子痫前期孕妇为病例组,并根据病情严重程度将病例组分为PE组(19例)和重度PE组(21例)。另选取同期在本院就诊的健康孕妇40例为对照组。分别采用RT-qPCR和免疫荧光法检测胎盘组织miR-210-5p和ITGAM表达。采用流式细胞术检测外周血单核细胞表面ITGAM表达。比较病例组与对照组临床资料以及ITGAM和miR-210-5p表达水平。比较不同病情严重程度PE患者ITGAM和miR-210-5p表达水平。分析miR-210-5p、ITGAM表达水平与PE患者临床资料的相关性。结果:病例组孕前体重指数(BMI)、收缩压、舒张压、尿酸(UA)、肌酐(Cr)、天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、乳酸脱氢酶(LDH)及平均血小板体积(MPV)高于对照组,分娩孕周、新生儿体重及白蛋白(ALB)低于对照组(均P<0.05)。与对照组比较,病例组外周血单核细胞表面以及胎盘组织ITGAM表达水平降低,胎盘组织miR-210-5p表达水平升高(均P<0.05)。与PE组比较,重度PE组胎盘组织ITGAM表达水平降低,miR-210-5p表达水平升高(均P<0.05)。胎盘组织miR-210-5p与ITGAM表达水平呈负相关(P<0.05)。胎盘组织miR-210-5p表达水平与患者收缩压、舒张压、AST、ALT、LDH及24 h尿蛋白定量呈正相关,与分娩孕周、新生儿体重及白蛋白水平呈负相关(均P<0.05)。胎盘组织ITGAM表达水平与患者收缩压、舒张压、AST、ALT、LDH及24 h尿蛋白定量呈负相关,与分娩孕周、新生儿体重及白蛋白水平呈正相关(均P<0.05)。外周血单核细胞表面ITGAM表达水平与患者LDH及24 h尿蛋白定量呈负相关,与白蛋白水平呈正相关(均P<0.05)。结论:PE孕妇胎盘组织miR-210-5p表达水平升高,外周血单核细胞表面及胎盘组织ITGAM表达水平均降低,且二者与PE的病情严重程度有关。
Abstract:
Objective:To investigate the expression levels and detection significance of microRNA-210-5p(miR-210-5p)and integrin αM(ITGAM)in pregnant women with preeclampsia(PE).Methods:A total of 40 pregnant women with preeclampsia were selected as the case group and divided into the PE group(19 cases)and severe PE group(21 cases)according to the severity of the condition.Another 40 healthy pregnant women during the same period served as the control group.The expression of miR-210-5p and ITGAM in placental tissue was detected by RT-qPCR and immunofluorescence,respectively.The expression of ITGAM on the surface of peripheral blood monocytes was detected by flow cytometry.Clinical data and expression levels of ITGAM and miR-210-5p were compared between the case and control groups.The expression levels of ITGAM and miR-210-5p in patients with different severity of PE were compared.The correlation between the expression levels of miR-210-5p and ITGAM and the clinical data of PE patients was analyzed.Results:The pre-pregnancy body mass index(BMI),systolic and diastolic blood pressure,uric acid(UA),creatinine(Cr),aspartate aminotransferase(AST),alanine aminotransferase(ALT),lactate dehydrogenase(LDH),and mean platelet volume(MPV)in the case group were higher than those in the control group,while the gestational age at delivery,newborn weight,and albumin(ALB)were lower(all P<0.05).Compared with the control group,the expression of ITGAM on the surface of peripheral blood monocytes and in placental tissue in the case group was decreased,while the expression of miR-210-5p in placental tissue was increased(all P<0.05).Compared with the PE group,the severe PE group had lower placental ITGAM expression and higher miR-210-5p expression(all P<0.05).The expression of miR-210-5p in placental tissue was negatively correlated with ITGAM expression(P<0.05).The expression of miR-210-5p in placental tissue was positively correlated with systolic and diastolic blood pressure,AST,ALT,LDH,and 24-hour urinary protein excretion,and negatively correlated with gestational age at delivery,newborn weight,and albumin levels(all P<0.05).The expression of ITGAM in placental tissue was negatively correlated with systolic and diastolic blood pressure,AST,ALT,LDH,and 24-hour urinary protein excretion,and positively correlated with gestational age at delivery,newborn weight,and albumin levels(all P<0.05).The expression of ITGAM on the surface of peripheral blood monocytes was negatively correlated with LDH and 24-hour urinary protein excretion,and positively correlated with albumin levels(all P<0.05).Conclusion:In pregnant women with preeclampsia,the expression of miR-210-5p in placental tissue is increased,and the expression of ITGAM on the surface of peripheral blood monocytes and in placental tissue is decreased,both of which are related to the severity of the condition.
