摘要:
目的:分析长链非编码RNA(lncRNA)TMPO-AS1通过磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶 B(Akt)信号通路对多囊卵巢综合征(PCOS)大鼠性激素和卵巢功能的影响。方法:随机将30只SD雌性大鼠分为模型组、si-NC组、TMPO-AS1-shRNA组,另选取健康大鼠设空白对照组,每组10只,模型组、si-NC组、TMPO-AS1-shRNA组给予60%高脂饲料,皮下注射DHEA(60 mg/kg溶于0.2 ml芝麻油中),空白对照组给予普通饲料,皮下注射等量芝麻油,于10 d开始对大鼠进行为期10 d的阴道涂片,持续注射21 d后测量大鼠体重、卵巢重量,计算卵巢指数; 并采用Western blot检测PI3K、P-PI3K、Akt、P-Akt蛋白水平; 采用荧光定量PCR(RT-qPCR)法检测TMPO-AS1、PI3K、Akt mRNA表达水平; 采用酶联免疫吸附法检测血清性激素水平; 于光学显微镜下观察卵巢形态学。结果:MPO-AS1、PI3K、Akt mRNA水平比较,模型组、si-NC组比空白对照组低(均P<0.05)。TMPO-AS1-shRNA组体重、卵巢重量、卵巢指数比空白对照组高,比模型组、si-NC组低(均P<0.05)。TMPO-AS1-shRNA组PI3K、Akt蛋白表达水平比空白对照组低,比模型组、si-NC组高(均P<0.05)。TMPO-AS1-shRNA组睾酮激素(T)、促黄体生成素(LH)水平比空白对照组高,比模型组、si-NC组低; 雌二醇(E2)、促卵泡生成激素(FSH)水平比空白对照组低,比模型组、si-NC组高(均P<0.05)。TMPO-AS1-shRNA组黄体数、成熟卵泡数、颗粒细胞厚层比空白对照组低,比模型组、si-NC组高(均P<0.05)。结论:lncRNA TMPO-AS1通过PI3K/Akt信号通路可有效降低PCOS大鼠性激素水平,并改善其卵巢功能。
Abstract:
Objective:To analyze the effects of long non-coding RNA(lncRNA)TMPO-AS1 on sex hormones and ovarian function in rats with polycystic ovary syndrome(PCOS)through phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt)signaling pathway.Methods:Thirty female SD rats were randomly divided into model group,si-NC group and TMPO-AS1-shRNA group,and 10 healthy rats were selected as blank control group.The model group,si-NC group and TMPO-AS1-shRNA group were fed with 60% high-fat diet,and DHEA(60 mg/kg dissolved in 0.2 ml sesame oil)was injected subcutaneously.The blank control group was given normal diet,and the same amount of sesame oil was injected subcutaneously.Vaginal smears were taken for 10 days from the 10th day,and the body weight and ovarian weight of the rats were measured after 21 days of continuous injection,and Ovarian Index was calculated.The protein levels of PI3K,p-PI3K,Akt and p-Akt were detected by Western blot.The mRNA levels of TMPO-AS1,PI3K and Akt were detected by RT-qPCR.Serum sex hormone levels were detected by enzyme-linked immunosorbent assay.Ovarian morphology was observed under an optical microscope.Results:The levels of MPO-AS1,PI3K and Akt mRNA in the model group and si-NC group were significantly lower than those in the blank control group(all P<0.05).The body weight,ovarian weight and Ovarian Index of TMPO-AS1-shRNA group were higher than those of blank control group,and lower than those of model group and si-NC group(all P<0.05).The protein expression levels of PI3K,Akt in TMPO-AS1-shRNA group were lower than those in blank control group,and higher than those in model group and si-NC group(all P<0.05).The levels of testosterone(T)and luteinizing hormone(LH)in TMPO-AS1-shRNA group were higher than those in blank control group,and lower than those in model group and si-NC group(all P<0.05).The levels of estradiol(E2)and follicle stimulating hormone(FSH)were lower than those in the blank control group,and higher than those in the model group and si-NC group(all P<0.05).The number of corpus luteum,the number of mature follicles and the thickness of granulosa cells in TMPO-AS1-shRNA group were lower than those in blank control group,and higher than those in model group and si-NC group(all P<0.05).Conclusion:lncRNA TMPO-AS1 can effectively reduce sex hormone levels and improve ovarian function in PCOS rats through PI3K/Akt signaling pathway.
