摘要:
目的:观察沙利度胺(THD)对类风湿关节炎(RA)成纤维样滑膜细胞(FLS)增殖的抑制作用及其对FLS和外周血单个核细胞(PBMCs)分泌肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)的影响,初步探讨THD治疗RA的可能机制。方法:分离RA患者FLS进行体外培养,采用四甲基偶氮唑蓝(MTT)法检测分别加入10、50、100、200 μg/ml的THD对FLS增殖的影响,采用酶联免疫吸附(ELISA)法检测50、100、200 μg/ml的THD分别对FLS及PBMCs进行干预24、48 h后分泌VEGF和TNF-α的变化。结果:THD在50～500 μg/ml浓度范围内对FLS增殖有抑制作用。与对照组比较,不同浓度(50、100、200 μg/ml)不同作用时间(分别干预24、48 h)的THD对FLS和PBMCs分泌TNF-α及VEGF均有抑制作用(均P<0.05)。结论:THD可能通过抑制RA患者 FLS的增殖,抑制FLS及PBMCs分泌VEGF、TNF-α,从而抑制新生血管的形成,减轻炎症反应,起到治疗RA的作用。
Abstract:
Objective:To observe the inhibitory effect of thalidomide(THD)on the proliferation of rheumatoid arthritis(RA)fibroblast-like synoviocytes(FLS)and its effect on the secretion of tumor necrosis factor-α(TNF-α)and vascular endothelial growth factor(VEGF)by FLS and peripheral blood mononuclear cells(PBMCs),and to explore the possible mechanism of THD in treating RA.Methods:FLS were separated and cultured in vitro.MTT method was applied to study the growth inhibition of FLS treated with different concentrations of THD(10,50,100,200 μg/ml).ELISA method was used to determine the expression levels of VEGF and TNF-α in FLS and PBMCs after 24 and 48 hours of intervention with different concentrations of THD(50,10,200 μg/ml).Results:THD inhibited the proliferation of FLS in the concentration range of 50 to 500 μg/ml.Compared with the control group,THD of different concentrations(50,100,200 μg/ml)and different duration(24 and 48 hours respectively)inhibited the secretion of TNF-α and VEGF by FLS and PBMCs(all P<0.05).Conclusion:THD may inhibit the proliferation of FLS and the secretion of VEGF and TNF-α by FLS and PBMCs in RA patients,thus inhibiting the formation of neovascularization and reducing inflammatory reaction,thus playing a role in the treatment of RA.
