摘要:
目的:探讨卵巢癌移植瘤裸鼠不同时期靶向超声造影参数与癌细胞生长的关系。方法:将人卵巢癌SKOV3细胞移植至50只BALB/c-Nude雌性裸鼠皮下制备卵巢癌移植瘤模型。在造模后第10、20、30、40、50天分别抽取10只裸鼠行超声造影检查。分析超声微泡物理性状及荧光鉴定结果,统计人卵巢癌移植造模结果,比较造模后不同时间点普通与靶向超声造影参数(峰值强度、达峰时间及平均渡越时间)。在不同时间点超声造影完成后处死裸鼠,剥离肿瘤,使用酶联免疫吸附试剂盒检测血管新生分子蛋白血管内皮生长因子受体1(VEGFR1)、酪氨酸激酶受体2(Tie-2)、细胞增殖蛋白[细胞周期蛋白D1(Cyclin D1)和E74样因子5(ELF5)]、细胞侵袭蛋白[胰岛素样生长因子结合蛋白2(IGFBP2)和E-上皮钙黏附素(E-cadherin)]水平。分析靶向超声造影参数与癌细胞生长的相关性。结果:靶向与普通微泡形态、大小比较均无明显差异。荧光显微镜下靶向造影微泡出现明亮红色荧光,普通造影微泡无荧光。造模后不同时间点,普通超声造影的峰值强度、达峰时间、平均渡越时间比较差异无统计学意义(均P>0.05); 而靶向超声造影的峰值强度、达峰时间、平均渡越时间比较差异有统计学意义(均P<0.05)。同一时间点时,靶向超声造影的峰值强度和平均渡越时间高于普通超声造影,达峰时间低于普通超声造影(均P<0.05)。造模后不同时间点,VEGFR1、Tie-2、Cyclin D1、ELF5、IGFBP2、E-cadherin相对表达量比较差异有统计学意义(均P<0.05)。Spearman相关性分析显示,靶向超声造影峰值强度及平均渡越时间与VEGFR1、Tie-2、Cyclin D1、IGFBP2、E-cadherin水平呈正相关,与ELF5呈负相关(均P<0.05); 达峰时间与VEGFR1、Tie-2、Cyclin D1、IGFBP2、E-cadherin水平呈负相关,与ELF5呈正相关(均P<0.05)。结论:不同时期超声靶向造影显影优于普通超声造影,超声造影参数与癌细胞生长有一定的相关性,有助于卵巢癌的早期诊断。
Abstract:
Objective:To investigate the relationship between of targeted contrast-enhanced ultrasound parameters at different stages and cancer cell growth in nude mice with ovarian cancer xenografts.Methods:Human ovarian cancer SKOV3 cells were transplanted into 50 BALB/c-nude female nude mice to prepare ovarian cancer xenograft model.Contrast-enhanced ultrasound was performed in 10 nude mice on days 10,20,30,40 and 50 after modeling,respectively.The physical characteristics and fluorescence identification results of ultrasound microbubbles were analyzed,and the results of human ovarian cancer transplantation were counted,and the common and targeted contrast-enhanced ultrasound parameters(peak intensity,peak time and average transit time)at different time points after modeling were compared.At different time points,the nude mice were killed and the tumor was dissected.Levels of angiogenic molecules VEGFR1,Tie-2,cell proliferation proteins(Cyclin D1 and ELF5),cell invasion proteins(IGFBP2 and E-cadherin)were measured by using enzyme-linked immunosorbent assays.The correlation between targeted contrast-enhanced ultrasound parameters and cancer cell growth was analyzed.Results:There was no difference in morphology and size between targeted microvesicles and ordinary microvesicles.The targeted contrast microbubbles showed bright red fluorescence under fluorescence microscope,while the ordinary contrast microbubbles showed no fluorescence.At different time points after modeling,there were no significant differences in peak intensity,peak time and average transit time of ordinary contrast-enhanced ultrasound(all P>0.05),while there were significant differences in peak intensity,peak time and average transit time of targeted contrast-enhanced ultrasound(all P<0.05).At the same time point,the peak intensity and average transit time of targeted contrast-enhanced ultrasound were higher than those of ordinary contrast-enhanced ultrasound,and the peak time was lower than that of ordinary contrast-enhanced ultrasound(all P<0.05).The relative expressions of VEGFR1,Tie-2,Cyclin D1,ELF5,IGFBP2 and E-cadherin were significantly different at different time points after modeling(all P<0.05).Spearman correlation analysis showed that peak intensity and average transit time of targeted contrast-enhanced ultrasound were positively correlated with VEGFR1,Tie-2,Cyclin D1,IGFBP2 and E-cadherin levels,and negatively correlated with ELF5(all P<0.05); peak time was negatively correlated with VEGFR1,Tie-2,Cyclin D1,IGFBP2 and E-cadherin levels,and positively correlated with ELF5(all P<0.05).Conclusion:Targeted contrast-enhanced ultrasound imaging at different stages is superior to ordinary contrast-enhanced ultrasound.Contrast-enhanced ultrasound parameters have certain correlation with cancer cell growth,which is helpful for early diagnosis of ovarian cancer.
