摘要:
铁死亡是近年来发现的一种铁依赖性的调节细胞死亡形式,其特征在于细胞内铁离子过载,活性氧自由基(ROS)和脂质过氧化物(LOOH)水平的显著性升高,进而引起细胞膜磷脂内侧发生严重的脂质过氧化反应,最终导致膜破裂和细胞崩解死亡。癫痫是神经系统常见的疾病之一,其特点为难治性、反复性和致残性,严重影响了患者的生活质量。铁代谢的异常、氧化应激、GPX4/GSH、脂氧合酶等因子介导的铁死亡,与癫痫的发病有着密切的联系,其所涉及的通路和机制处于不断明确中,且铁死亡抑制剂能在一定程度上阻止癫痫的发作及发展; 同时铁死亡研究很有可能成为一个新的支点,在癫痫的预防及治疗上发挥至关重要的作用。现就铁死亡相关通路与癫痫关系研究进展综述如下。
Abstract:
Ferroptosis was discovered in recent years,a form of iron dependency regulation of cell death,the characteristic is iron overload in the cell,active oxygen free radicals(ROS)and lipid peroxide(LOOH)levels significantly increased,thus causing serious lipid peroxidation of cell membrane phospholipids inside,eventually leading to membrane rupture and cell disintegration and death.Epilepsy is one of the common diseases of nervous system,which is characterized by refractory,recurrent and disabling,and seriously affects the quality of life of patients.Ferroptosis mediated by abnormal iron metabolism,oxidative stress,GPX4/GSH,lipoxygenase and other factors is closely related to the pathogenesis of epilepsy.The pathways and mechanisms involved are constantly being clarified,and ferroptosis inhibitors can prevent the onset and development of epilepsy to a certain extent.At the same time,ferroptosis research is likely to become a new fulcrum and play a crucial role in the prevention and treatment of epilepsy.The research progress of Ferroptosis-related pathways and epilepsy is reviewed as follows.
