摘要:
目的:研究桃叶珊瑚苷在体外对白细胞介素-1β(IL-1β)诱导的软骨细胞凋亡的影响,并探讨桃叶珊瑚苷在体内对小鼠骨关节炎(OA)疾病进展的影响。方法:①将小鼠软骨细胞分为对照组、IL-1β组和IL-1β+AC组。对照组小鼠软骨细胞正常培养36 h。IL-1β组小鼠软骨细胞正常培养12 h后,加入10 μg/L IL-1β培养24 h。IL-1β+AC组小鼠软骨细胞加入100 μmol/L桃叶珊瑚苷处理12 h后,加入10 μg/L IL-1β培养24 h。比较三组软骨细胞活性、凋亡率、凋亡相关基因(Bax和Bcl-2)以及炎症因子[肿瘤坏死因子(TNF-α)、IL-6、前列腺素E2(PGE2)和基质金属蛋白酶-3(MMP-3)]表达情况。②将30只C57小鼠随机分为正常组、模型组和AC组。模型组和AC组小鼠经手术诱导建立OA模型。AC组小鼠按照80 mg/(kg·d)治疗6周,其他两组小鼠给予等量0.9%氯化钠溶液。比较三组小鼠国际骨关节炎研究学会(OARSI)评分、软骨下骨骨量、软骨组织中细胞凋亡率以及血清炎症因子含量。结果:①与对照组相比,IL-1β组软骨细胞活性显著下降,凋亡率显著上升,经桃叶珊瑚苷处理后IL-1β+AC组软骨细胞活性升高,凋亡率下降(均P<0.05)。与对照组相比,IL-1β组和IL-1β+AC组软骨细胞内Bax mRNA表达显著升高,而Bcl-2 mRNA表达显著降低(均P<0.05)。与IL-1β组相比,IL-1β+AC组软骨细胞内Bax mRNA表达显著降低,而Bcl-2 mRNA表达显著升高(均P<0.05)。与对照组相比,IL-1β组和IL-1β+AC组软骨细胞内TNF-α、IL-6、PGE2和MMP-3含量显著升高,且IL-1β+AC组软骨细胞内TNF-α、IL-6、PGE2和MMP-3含量显著低于IL-1β组(均P<0.05)。②与正常组小鼠相比,模型组和AC组小鼠软骨OARSI评分、软骨下骨骨量及软骨组织内细胞凋亡率显著升高,且AC组小鼠软骨OARSI评分、软骨下骨骨量与软骨组织内细胞凋亡率显著低于模型组(均P<0.05)。与正常组相比,模型组和AC组血清TNF-α、IL-6、PGE2和MMP-3含量显著升高,且AC组血清TNF-α、IL-6、PGE2和MMP-3含量显著低于模型组(均P<0.05)。结论:桃叶珊瑚苷在体外显著抑制IL-1β诱导的软骨细胞凋亡,并在体内延缓小鼠OA疾病进展,其机制可能与桃叶珊瑚苷通过抑制炎症因子表达而抑制炎症诱导的软骨细胞凋亡有关。
Abstract:
Objective:To study study the effect of aucubin on the apoptosis of chondrocytes induced by interleukin-1β(IL-1β)in vitro,and to explore the effect of aucubin on the progression of osteoarthritis(OA)in mice in vivo.Methods:①Mouse chondrocytes were divided into control group,IL-1β group and IL-1β+AC group.Chondrocytes in control group were routinely cultured for 36 hours.Chondrocytes in IL-1β group were routinely cultured for 12 hours,and 10 μg/L IL-1β was added for 24 hours.Chondrocytes in IL-1β+AC group were treated with 100 μmol/L aucubin for 12 hours,and cultured with 10 μg/L IL-1β for 24 hours.Chondrocyte activity,apoptosis rate,expression of apoptosis-related genes(Bax and Bcl-2)and inflammatory factors(TNF-α,IL-6,PGE2 and MMP-3)were compared among the three groups.②Thirty C57 mice were randomly divided into normal group,model group and AC group.OA model was established by surgical induction in mice of model group and AC group.Mice in the AC group were treated with 80 mg/(kg·d)for 6 weeks,and the other two groups were given the same amount of 0.9% sodium chloride solution.The OARSI score,subchondral bone mass and apoptosis rate in cartilage tissue,and serum inflammatory factors were compared among the three groups.Results:①Compared with the control group,the activity of chondrocytes in IL-1β group was significantly decreased and the apoptosis rate was significantly increased,and the activity of chondrocytes in IL-1β+AC group was increased and the apoptosis rate was decreased after treatment with aucubin(all P<0.05).Compared with the control group,the expression of Bax mRNA in chondrocytes in IL-1β group and IL-1β+AC group was significantly increased,while the expression of Bcl-2 mRNA was significantly decreased(all P<0.05).Compared with IL-1β group,the expression of Bax mRNA in chondrocytes of IL-1β+AC group was significantly decreased,while the expression of Bcl-2 mRNA was significantly increased(all P<0.05).Compared with the control group,the contents of TNF-α,IL-6,PGE2 and MMP-3 in chondrocytes of IL-1β group and IL-1β+AC group were significantly increased,and those in chondrocytes of IL-1β+AC group were significantly lower than those of IL-1β group(all P<0.05).②Compared with normal group,the OARSI score,subchondral bone mass and apoptosis rate of cartilage tissue in model group and AC group were significantly increased,and those of cartilage tissue in AC group were significantly lower than those in model group(all P<0.05).Compared with the normal group,the levels of TNF-α,IL-6,PGE2 and MMP-3 in the model group and AC group were significantly increased,and those in the AC group were significantly lower than those in the model group(all P<0.05).Conclusion:Aucubin can significantly inhibit IL-1β-induced chondrocyte apoptosis in vitro and delay the progression of OA disease in mice in vivo.The mechanism may be related to the inhibition of inflammation-induced chondrocyte apoptosis by the inhibition of the expression of inflammatory factors.
