摘要:
目的:观察替格瑞洛对脓毒血症小鼠高迁移率蛋白1(HMGB-1)及免疫功能的影响。方法: 将100只同月龄健康SPF级Wistar小鼠,随机选取80只小鼠进行脓毒血症造模将其依次设为对照组及替格瑞洛低、中、高剂量组,每组各20只; 将20只未制备脓毒血症小鼠模型的小鼠设为空白组。观察各组小鼠存活率、脾指数、谷丙转氨酶(GPT)、谷草转氨酶(GOT)、免疫功能、Bcl-2相关X蛋白(Bax)、B细胞淋巴瘤-2(Bcl-2)、HMGB-1蛋白表达。结果: 空白组存活率、脾指数、Bax、Bcl-2、GPT、GOT、CD4+、CD8+、Th1细胞(Th1)、Th2细胞(Th2)、T细胞17(Th17)、调节性T细胞(Treg)、HMGB-1蛋白水平优于对照组、替格瑞洛低、中、高剂量组,替格瑞洛高剂量组存活率、脾指数、Bax、Bcl-2、GPT、GOT、CD4+、CD8+、Th1、Th2、Th17、Treg、HMGB-1蛋白水平优于对照组、替格瑞洛低、中剂量组,替格瑞洛中剂量组存活率、脾指数、Bax、Bcl-2、GPT、GOT、CD4+、CD8+、Th1、Th2、Th17、Treg、HMGB-1蛋白水平优于对照组、替格瑞洛低剂量组,替格瑞洛低剂量组存活率、脾指数、Bax、Bcl-2、GPT、GOT、CD4+、CD8+、Th1、Th2、Th17、Treg、HMGB-1蛋白水平优于对照组,差异有统计学意义(均P<0.05)。结论: 替格瑞洛可通过调节HMGB-1表达而恢复免疫功能,进而抑制炎症因子释放,而避免机体免疫损伤。
Abstract:
Objective:To observe the effect of ticagrelor on high mobility protein 1(HMGB-1)and immune function in septic mice.Methods:A total of 100 healthy SPF Wistar mice of the same month were selected as the research subjects.Eighty mice were randomly selected for sepsis modeling and divided into control group and ticagrelor low-dose group,ticagrelor medium-dose group and ticagrelor high-dose group,with 20 mice in each group.Twenty mice without sepsis model were set as blank group.The survival rate,Spleen index,and the levels of GPT,GOT,immune function indexes,Bax,Bcl-2 and HMGB-1 proteins were observed.Results:The survival rate,Spleem index,and levels of Bax,Bcl-2,GPT,GOT,CD4+,CD8+,Th1,Th2,Th17,Treg and HMGB-1 protein in the blank group were better than those in other groups(all P<0.05).The above indexes in the ticagrelor high-dose group were better than those in the control group,ticagrelor low- and medium-dose groups(all P<0.05).The above indexes in the medium-dose group were better than those in the control group and ticagrelor low-dose group(all P<0.05).The above indexes in the ticagrelor low-dose group were significantly better than those in the control group(all P<0.05).Conclusion:Ticagrelor can restore immune function by regulating the expression of HMGB-1,thereby inhibiting the release of inflammatory factors and avoiding immune damage.
