摘要:
目的:探讨半乳糖凝集素-3抑制剂柑橘果胶对心力衰竭大鼠血浆N端B型利钠肽原(NT-proBNP)及心肌纤维化的影响。方法:将心力衰竭大鼠(42只)随机平分为模型组、柑橘果胶A组和柑橘果胶B组。模型组给予10 mg/(kg·d)的0.9%氯化钠溶液灌胃。柑橘果胶A组、B组分别给予0.10、0.20 mg/(kg·d)的柑橘果胶加入0.9%氯化钠溶液灌胃,连续治疗并观察4周。检测三组大鼠血浆NT-proBNP表达及心肌纤维化的变化情况。结果:在治疗第2、4周,柑橘果胶A组、B组室间隔厚度(IVS)高于模型组,左心室舒张末期内径(LVDD)低于模型组,且柑橘果胶B组与柑橘果胶A组比较差异有统计学意义(均P<0.05)。在治疗第4、8周,柑橘果胶A组和B组NT-proBNP、心脏指数、心肌胶原容积分数以及Ⅰ型和Ⅲ型胶原蛋白相对表达水平低于模型组,且柑橘果胶B组上述指标低于柑橘果胶A组(均P<0.05)。结论:半乳糖凝集素-3抑制剂柑橘果胶能够抑制心力衰竭大鼠血浆NT-proBNP的表达,改善心肌纤维化,进而改善心脏指数、心肌胶原容积分数以及心脏结构形态。
Abstract:
Objective:To investigate the effects of Galectin-3 inhibitor citrus pectin on plasma N-terminal pro-B-type natriuretic peptide(NT-proBNP)and myocardial fibrosis in rats with heart failure.Methods: Rats with heart failure(n=42)were randomly divided into model group,citrus pectin group A,citrus pectin group B,with 14 rats in each group.The model group was given 10 mg/(kg·d)of 0.9% sodium chloride solution by gavage.Citrus pectin group A and group B were treated with 0.10 mg/(kg·d)and 0.20 mg/(kg·d)of citrus pectin by gavage with 0.9% sodium chloride solution for 4 weeks.The changes of plasma NT-proBNP expression and myocardial fibrosis in the three groups of rats were detected.Results:In the 2nd and 4th week of treatment,IVS of citrus pectin group A and group B was higher than that of model group,LVDD was lower than that of model group,and the difference between citrus pectin group B and citrus pectin group A was statistically significant(all P<0.05).In 4th and 8th week of treatment,NT-proBNP,cardiac index,cardiac collagen volume fraction and relative expression levels of type Ⅰ and type Ⅲ collagen in citrus pectin group A and group B were lower than those in model group,and the above indexes in citrus pectin group B were lower than those in citrus pectin group A(all P<0.05).Conclusion:Galectin-3 inhibitor citrus pectin can inhibit the expression of plasma NT-proBNP in rats with heart failure,improve myocardial fibrosis,and further improve cardiac index,myocardial collagen volume fraction and cardiac structure.
