摘要:
目的:探讨骨桥蛋白(OPN)基因沉默对子宫内膜异位症(EMS)患者在位内膜细胞侵袭性的影响。方法:采用原代细胞培养方法培养EMS患者在位内膜腺上皮细胞,分为干预组和未干预组。干预组采用OPN特异小RNA(siRNA)进行基因沉默。采用Western blot、RT-PCR检测细胞中OPN蛋白、mRNA表达情况。采用穿膜实验观察细胞的侵袭性变化。结果:与未干预组比较,干预组细胞OPN蛋白及mRNA表达明显下降(均P<0.05)。与未干预组比较,干预组细胞穿膜数减少(P<0.05)。结论:对EMS患者在位内膜腺上皮细胞进行OPN基因沉默后,细胞OPN蛋白、mRNA表达降低,侵袭性明显减弱,说明OPN与细胞侵袭性明显相关。
Abstract:
Objective:To investigate the effect of osteopontin(OPN)gene silencing on the invasiveness of eutopic endometrial cells in patients with endometriosis(EMS).Methods: The epithelial cells of eutopic endometrium from EMS patients were cultured by primary cell culture and divided into intervention group and non-intervention group.In the intervention group,OPN interfering small RNA(siRNA)was used for gene silencing.Western blot and RT-PCR were used to detect the expression of OPN protein and mRNA in cells.The change of cellular invasiveness was observed by transmembrane test.Results:Compared with the non-intervention group,the expressions of OPN protein and mRNA in the intervention group was significantly decreased(all P<0.05).Compared with the non-intervention group,the number of cells penetrating the membrane decreased in the intervention group(P<0.05).Conclusion:After OPN gene silencing of EMS glandular epithelial cells,the expressions of OPN protein and mRNA decreased,and the invasiveness decreased,which indicated that OPN was significantly related to cell invasiveness.
