摘要:
目的:探讨右美托咪定对慢性心力衰竭大鼠的作用及机制。方法:将慢性心力衰竭大鼠(42只)随机平分为模型组、右美托咪定A组与右美托咪定B组。三组大鼠分别给予0、5、10 μg/kg右美托咪定腹腔注射。观察大鼠认知功能变化情况,检查大鼠左心室射血分数(LVEF)。处死大鼠后,取心脏称重并计算心脏质量指数,检测大鼠血清神经元特异烯醇酶(NSE)与超氧化物歧化酶(SOD)活性,检测环磷腺苷效应元件结合蛋白(CREB)/脑源性神经营养因子(BNDP)通路相关蛋白表达变化情况。结果:右美托咪定A组与右美托咪定B组给药后第7、14天每分钟穿越平台次数多于模型组,逃避潜伏期低于模型组(均P<0.05)。右美托咪定B组给药后第7、14天每分钟穿越平台次数多于右美托咪定A组,逃避潜伏期低于右美托咪定A组(均P<0.05)。右美托咪定A组与右美托咪定B组给药后第14天LVEF高于模型组,心脏质量指数低于模型组(均P<0.05)。右美托咪定B组给药后第14天LVEF高于右美托咪定A组,心脏质量指数低于右美托咪定A组(均P<0.05)。右美托咪定A组与右美托咪定B组给药后第14天血清NSE含量低于模型组,血清SOD活性高于模型组(均P<0.05)。右美托咪定B组血清NSE含量低于右美托咪定A组,血清SOD活性高于右美托咪定A组(均P<0.05)。右美托咪定A组与右美托咪定B组给药后第14天心肌组织CREB与BDNF蛋白相对表达水平高于模型组(均P<0.05)。右美托咪定B组给药后第14天心肌组织CREB与BDNF蛋白相对表达水平高于右美托咪定A组(均P<0.05)。结论:右美托咪定能提高慢性心力衰竭大鼠心肌组织CREB与BDNF蛋白水平,抑制NSE释放,提高SOD活性,改善大鼠的心功能,还可促进认知功能的恢复。
Abstract:
Objective:To investigate the effect and mechanism of dexmedetomidine on chronic heart failure in rats.Methods: Rats with chronic heart failure(n=42)were randomly divided into model group,dexmedetomidine group A and dexmedetomidine group B.The rats in the three groups were given 0,5 and 10 μg/kg dexmedetomidine intraperitoneally.The changes of cognitive function and left ventricular ejection fraction(LVEF)were observed.After the rats were killed,the heart was weighed and the cardiac mass index was calculated,and the serum NSE content and SOD activity were detected,and the expression of CREB /BNDP pathway related proteins were detected.Results:On the 7th and 14th days after administration,dexmedetomidine group A and dexmedetomidine group B had more times to cross the platform per minute than the model group,and the escape latency was lower than that of the model group(all P<0.05); dexmedetomidine group B had more times to cross the platform per minute than dexmedetomidine group A,and the escape latency was lower than that of dexmedetomidine group A(all P<0.05).On the 14th day after administration,LVEF in dexmedetomidine group A and dexmedetomidine group B was higher than that in model group,and cardiac mass index was lower than that in model group(all P<0.05); LVEF in dexmedetomidine group B was higher than that in dexmedetomidine group A,and cardiac mass index was lower than that in dexmedetomidine group A(all P<0.05).On the 14th day after administration,serum NSE content of dexmedetomidine group A and dexmedetomidine group B was lower than that of model group,and serum SOD activity was higher than that of model group(all P<0.05); serum NSE content of dexmedetomidine group B was lower than that of dexmedetomidine group A,and the serum SOD activity was higher than that of dexmedetomidine group A(all P<0.05).On the 14th day after administration,the relative expression levels of CREB and BDNF proteins in myocardial tissue of dexmedetomidine group A and dexmedetomidine group B were higher than those of model group(all P<0.05); the relative expression levels of CREB and BDNF proteins in myocardial tissue of dexmedetomidine group B were higher than those of dexmedetomidine group A(all P<0.05).Conclusion:Dexmedetomidine can increase the levels of CREB and BDNF proteins,inhibit NSE release,increase SOD activity,improve the cardiac function of rats with chronic heart failure,and promote the recovery of cognitive function.
