摘要:
目的:探讨阿比多尔治疗慢性阻塞性肺疾病(COPD)大鼠的效果并分析对大鼠气道炎症和气道黏液高分泌相关指标的影响。方法:将45只健康雄性SD大鼠依据随机数字表法分为对照组、模型组和阿比多尔组,各15只。除对照组外,其余两组均采用烟熏和气管内注射脂多糖法建立COPD模型。造模成功后,阿比多尔组大鼠用3 g/L阿比多尔溶液 1 ml/100 g灌胃; 对照组和模型组用生理盐水1 ml/100 g灌胃,三组连续灌胃10 d。使用动物肺功能仪测定各组大鼠肺活量(FVC)、呼气峰值流速(PEF)、第0.1 s用力呼气容积(FEV0.1)、气道阻力(RI)等肺功能指标。采用酶联免疫吸附法检测各组大鼠支气管肺泡灌洗液中肿瘤坏死因子(TNF-α)、白细胞介素-8(IL-8)、IL-17等炎症因子水平。分别采用Western blot技术和实时荧光定量PCR技术检测各组大鼠支气管肺组织中黏蛋白5AC(MUC5AC)和钙离子激活的氯离子通道1(CLCA1)和Toll 样受体4蛋白(TLR4)表达情况。结果:阿比多尔组大鼠FVC、PEF、FEV0.1均显著高于模型组,但显著低于对照组; RI显著低于模型组,但显著高于对照组(均P<0.05)。阿比多尔组大鼠支气管肺泡灌洗液TNF-α、IL-8、IL-17水平均显著高于对照组,但显著低于模型组(均P<0.05)。阿比多尔组大鼠肺组织MUC5AC、CLCA1和TLR4 蛋白相对吸光值和mRNA相对表达量均显著低于模型组,但显著高于对照组(均P<0.05)。结论:阿比多尔能够显著改善COPD大鼠肺功能,这与阿比多尔减轻气道炎症和气道黏液高分泌有关。
Abstract:
Objective:To investigate the effect of abidol on chronic obstructive pulmonary disease(COPD)rats and to analyze the effects on airway inflammation and airway mucus hypersecretion in rats.Methods:45 healthy male SD rats were randomly divided into control group,model group and abidol group,with 15 cases in each group.Except the control group,the COPD models were established by smoking and intratracheal injection of lipopolysaccharide.After successful modeling,rats in abidol group were gavaged with abidol solution(3 g/L)1 ml/100 g,and the control group and model group were gavaged with normal saline 1 ml/100 g,and the three groups were gavaged continuously for 10 days.The lung function indexes such as forced vital capacity(FVC),peak expiratory flow rate(PEF),forced expiratory volume in 0.1 second(FEV0.1)and airway resistance(RI)were measured by animal lung function instrument.The levels of tumor necrosis factor-α(TNF-α),interleukin-8(IL-8)and IL-17 in bronchoalveolar lavage fluid were detected by enzyme-linked immunosorbent assay.The expressions of mucin 5AC(MUC5AC),calcium activated chloride channel 1(CLCA1)and toll like receptor 4 protein(TLR4)were detected by Western blot and real-time fluorescence quantitative PCR.Results:FVC,PEF and FEV0.1 in the abidol group was significantly higher than those in the model group,but significantly lower than those in the control group(all P<0.05).RI in the abidol group was significantly lower than that in the model group,but significantly higher than that in the control group(all P<0.05).The levels of TNF-α,IL-8 and IL-17 in bronchoalveolar lavage fluid of abidol group were significantly higher than thoset of control group,but significantly lower than those of model group(all P<0.05).The relative light absorption value of MUC5AC,CLCA1 and TLR4 proteins and the relative expression levels of MUC5AC,CLCA1 and TLR4 mRNAs in the abidol group were significantly lower than those in the model group,but significantly higher than those in the control group(all P<0.05).Conclusion:Abidol can significantly improve pulmonary function in COPD rats,which is related to abidor's reduction of airway inflammation and airway mucus hypersecretion.
