摘要:
目的:探讨戊巴比妥钠诱导心力衰竭相关的H9c2大鼠心肌细胞损伤模型的制作并明确相关机制。方法:不同浓度不同造模时间戊巴比妥钠对H9c2心肌细胞进行干预,利用透射电镜检测细胞超微结构、三磷酸腺苷(ATP)、活性氧(ROS)、细胞凋亡、自噬等细胞改变。结果:模型组(戊巴比妥钠干预组)线粒体可见肿胀边缘不清、内膜破裂、嵴断裂等改变,并伴随自噬小体增多。模型组ATP含量显著减少(P<0.01),ROS水平明显增加,细胞凋亡率显示出增长趋势(P>0.05)。结论:戊巴比妥钠诱导的H9c2心肌细胞损伤模型符合心衰心肌细胞病理改变,明确戊巴比妥钠对心肌细胞的影响,可为体外心衰模型的研究提供基础理论。
Abstract:
Objective:To explore the establlshment of pentobarbital sodium induced heart failure cardiomyocytes injury model of rat H9c2 cells and the related mechanism.Methods:Different concentrations and modeling time of pentobarbital sodium intervened H9c2 cardiomyocytes.The changes of cell ultrastructure,ATP,reactive oxygen species(ROS),apoptosis and autophagy were detected by transmission electron microscope.Results:In the model group(pentobarbital sodium intervention group),mitochondria showed swelling,unclear edge,intimal rupture,ridge rupture and other changes,accompanied by the increase of autophagy bodies.In the model group,the content of ATP decreased significantly(P<0.01),the level of ROS increased significantly,and the apoptosis rate showed an increasing trend(P>0.05).Conclusion:The H9c2 cardiomyocytes injury model induced by pentobarbital sodium is consistent with the pathological changes of cardiomyocytes in heart failure.Clarifying the effect of pentobarbital sodium on cardiomyocytes can provide a basic theory for the study of heart failure model in vitro.
