摘要:
目的:探究右美托咪定对坐骨神经慢性压迫损伤(CCI)模型大鼠神经病理性疼痛的缓解作用及机制。方法:选择100只雄性SD大鼠,随机分为空白对照组、模型组、右美托咪定低剂量组、右美托咪定中剂量组和右美托咪定高剂量组。除空白对照组外,其他四组均构建坐骨神经CCI模型。比较各组大鼠海马区高迁移率组蛋白1(HMGB1)、肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)表达水平,以及HMGB1及其下游Toll样受体4(TLR4)和糖基化终产物受体(RAGE)mRNA相对表达水平。结果:与空白对照组比较,模型组大鼠海马区HMGB1蛋白相对表达量、TNF-α和IL-1β表达水平、HMGB1及其下游TLR4和RAGE mRNA相对表达水平表达较高(均P<0.05)。与模型组比较,右美托咪定各剂量组上述指标均降低(均P<0.05)。结论:右美托咪定可以减轻坐骨神经CCI模型大鼠神经病理性疼痛,其机制可能与抑制HMGB1从而抑制星形胶质细胞活性和TLR4/核因子-κB信号通路有关。
Abstract:
Objective:To explore the alleviating effect and mechanism of dexmedetomidine on chronic neuropathic pain in rat model with chronic compression injury(CCI)of sciatic nerve.Methods:A total of 100 male SD rats were selected and randomly divided into blank control group,model group,low-dose dexmedetomidine group,middle-dose dexmedetomidine group,and high-dose dexmedetomidine group.Except for the blank control group,the sciatic nerve CCI model was constructed in the other four groups.The relative expression levels of high mobility histone 1(HMGB1),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in the hippocampus of each group of rats,as well as the relative expression levels of HMGB1 and its downstream Toll-like receptor 4(TLR4)and receptor for advanced glycation endproduct(RAGE)mRNA were compared between groups.Results:Compared with the blank control group,the relative expression levels of HMGB1 protein,TNF-α and IL-1β,and the relative expression levels of HMGB1 and its downstream TLR4 and RAGE mRNA in the hippocampus of the model group were higher(all P<0.05).Compared with the model group,the above indicators in each dose group of dexmedetomidine were decreased(all P<0.05).Conclusion:Dexmedetomidine can alleviate neuropathic pain in sciatic nerve CCI model rats,and its mechanism may be related to the inhibition of HMGB1,thereby inhibiting the activity of astrocytes and the TLR4/NF-κB signaling pathway.
