摘要:
目的:探究miRNA-26a-5p靶向调控蛋白酪氨酸磷酸酶基因(PTEN)对冠心病(CHD)模型大鼠主动脉内皮细胞炎症损伤的影响。方法:选取健康SD大鼠22只,随机分成正常组和模型组,正常组大鼠不用进行特殊处理,模型组大鼠按高脂饲料喂养法制备成冠心病模型,对比两组大鼠血钙和血脂水平、大鼠主动脉内皮组织的miRNA-26-5p、PTEN、caspase-1 mRNA相对表达量及PTEN、caspase-1蛋白相对表达量。取模型组大鼠剩余的部分主动脉内皮组织进行细胞培养,经细胞转染后,分成空白对照组、阴性组、miRNA-26a-5p mimic组、miRNA-26a-5p inhibitor组和miRNA-26a-5p inhibitor+siPTEN组,对比五组的miRNA-26a-5p、PTEN蛋白,miRNA-26-5p、PTEN、caspase-1 mRNA相对表达量和4个炎症因子(IL-10、IL-1β、IL-6和IL-18)水平。结果:①与正常组对比,模型组的总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDLC)、低密度脂蛋白胆固醇(LDLC)和Ca2+水平均高于正常组(均P<0.05)。②模型组主动脉内皮组织miRNA-26a-5p水平低于正常组,PTEN、caspase-1 mRNA表达水平以及PTEN、caspase-1蛋白表达水平均高于正常组(均P<0.05)。③与空白对照组对比,miRNA-26a-5p mimic组miRNA-26a-5p相对表达升高,PTEN、caspase-1 mRNA相对表达降低,PTEN、caspase-1蛋白表达升高(均P<0.05); miRNA-26a-5p inhibitor组的miRNA-26a-5p相对表达降低,PTEN、caspase-1 mRNA相对表达升高,PTEN、caspase-1蛋白表达降低(均P<0.05)。与miRNA-26a-5p mimic组相比,miRNA-26a-5p inhibitor+siPTEN组的miRNA-26a-5p相对表达降低,PTEN、caspase-1 mRNA相对表达均升高,PTEN、caspase-1蛋白表达均降低(均P<0.05)。④与空白对照组、阴性组相比,miRNA-26a-5p mimic组IL-1β、IL-6和IL-18水平降低,IL-10升高(均P<0.05),miRNA-26a-5p inhibitor组与之相反(P<0.05); 与miRNA-26a-5p mimic组相比,miRNA-26a-5p inhibitor组IL-1β、IL-6和IL-18水平升高,IL-10降低(均P<0.05)。结论:冠心病大鼠主动脉内皮细胞中miRNA-26a-5p呈高表达、PTEN低表达,miRNA-26a-5p靶向调控PTEN能够使IL-1β、IL-6和IL-18炎症因子释放减少,减缓主动脉内皮细胞炎症损伤,具有内皮细胞保护作用。
Abstract:
Objective:To investigate the effect of targeted regulation of protein tyrosine phosphatase gene(PTEN)by miRNA-26a-5p on inflammatory injury of aortic endothelial cells in rats with coronary heart disease(CHD).Methods:22 healthy SD rats were randomly divided into normal group and model group.The rats in the normal group did not need special treatment.The rats in the model group were fed with high-fat diet to prepare the coronary heart disease model.The blood calcium and blood lipid levels,the expression levels of miRNA-26-5p,PTEN,caspase-1 mRNAs,and the expression levels of PTEN and caspase-1 proteins in aortic endothelium of the two groups were compared.Part of the aortic endothelial tissue of the model group was cut for cell culture.After cell transfection,the cells were divided into blank control group,negative control group,miRNA-26a-5p mimic group,miRNA-26a-5p inhibitor group and miRNA-26a-5p inhibitor + siPTEN group.The miRNA-26a-5p,PTEN mRNA,PTEN protein,caspase-1 mRNA,caspase-1 protein and four inflammatory factors(IL-10,IL-1,IL-6 and IL-18)in the five groups were compared.Results:①Compared with the normal group,the levels of TC,TG,LDLC and Ca2+ in the model group were significantly higher than those in the normal group(all P<0.05).②The level of miRNA-26a-5p in the model group was significantly lower than that in the normal group,and the expression levels of PTEN mRNA,caspase-1 mRNA,PTEN protein and caspase-1 protein were significantly higher than those in the normal group(all P<0.05).③Compared with the blank control group,the relative expression of miRNA-26a-5p increased significantly,the relative expression of PTEN and caspase-1 mRNAs decreased significantly,and the expression of PTEN and caspase-1 proteins increased significantly(all P<0.05).In the miRNA-26a-5p inhibitor group,the relative expression of miRNA-26a-5p decreased significantly,the relative expression of PTEN and caspase-1 mRNAs increased,and the expression of PTEN and caspase-1 proteins decreased(all P<0.05).Compared with miRNA-26a-5p mimic group,the relative expression of miRNA-26a-5p in miRNA-26a-5p inhibitor + siPTEN group decreased,the relative expression of PTEN and caspase-1 mRNAs increased,and the expression of PTEN and caspase-1 proteins decreased(all P<0.05).④Compared with blank control group and negative control group,the levels of IL-1β,IL-6 and IL-18 in miRNA-26a-5p mimic group decreased significantly,and IL-10 increased significantly,which was opposite in miRNA-26a-5p inhibitor group(all P<0.05); Compared with miRNA-26a-5p mimic group,miRNA-26a-5p inhibitor group,the levels of IL-1β,IL-6 and IL-18 increased significantly,and IL-10 decreased significantly(all P<0.05).Conclusion:MiRNA-26a-5p was highly expressed and PTEN was lowly expressed in aortic endothelial cells of rats with coronary heart disease.MiRNA-26a-5p targeting PTEN can increase IL-1β,IL-6 and IL-18,reduce the release of inflammatory factors,slow down the inflammatory injury of aortic endothelial cells,and have the protective effect of endothelial cells.
