摘要:
目的:探讨血管生成素-2(Ang-2)通过整合素αvβ1对内皮细胞基质金属蛋白酶-2(MMP-2)表达的影响。方法:体外培养人脐静脉血管内皮细胞,将细胞接种于12孔培养板中,当细胞汇合度达到80%左右时,终止消化,分为对照组、空腺病毒转染组、Ang-2腺病毒转染组和αvβ1 siRNA+Ang-2腺病毒转染组。每组设6个复孔,将培养板置于培养箱中72 h。用Western blot法检测Ang-2、αvβ1和MMP-2蛋白表达水平。结果:空腺病毒转染组Ang-2、αvβ1和MMP-2表达水平较对照组无统计学差异(均P>0.05)。Ang-2腺病毒转染组Ang-2和MMP-2表达水平较对照组均明显升高(均P<0.05),αvβ1表达水平较对照组无统计学差异(P>0.05)。αvβ1 siRNA+Ang-2腺病毒转染组Ang-2表达水平较对照组明显升高(P<0.05),但较Ang-2腺病毒转染组无统计学差异(P>0.05),αvβ1表达水平较Ang-2腺病毒转染组明显下降(P<0.05),MMP-2表达水平较Ang-2腺病毒转染组也明显下降(P<0.05)。结论:Ang-2可能通过结合αvβ1促进内皮细胞MMP-2表达。
Abstract:
Objective:To investigate the effect of angiopoietin-2(Ang-2)on the expression of matrix metalloproteinase-2(MMP-2)in endothelial cells through integrin αVβ1.Methods:Human umbilical vein endothelial cells(HUVECs)were cultured in vitro.The cells were inoculated into 12 well culture plates.When the cell confluence reached 80%,the digestion was terminated.The cells were divided into control group,empty adenovirus transfection group,Ang-2 adenovirus transfection group and αvβ1 siRNA+Ang-2 adenovirus transfection group.Each group had 6 multiple wells,and the culture plate was placed in the incubator for 72 hours.Western blot was used to detect the levels of Ang-2,αvβ1 and MMP-2 proteins.Results:The levels of Ang-2,αvβ1 and MMP-2 proteins in empty adenovirus transfection group were not significantly different from control group(all P>0.05).The levels of Ang-2 and MMP-2 protein in Ang-2 adenovirus transfection group were significantly higher than those in control group(all P<0.05),but there was no significant difference in the expression level of αvβ1 between Ang-2 adenovirus transfection group and control group(P>0.05).The expression of Ang-2 protein in αvβ1 siRNA+Ang-2 adenovirus transfection group was significantly higher than that in control group(P<0.05),but there was no significant difference compared with Ang-2 adenovirus transfection group(P>0.05).The αvβ1 and MMP-2 expression levels in αvβ1 siRNA+Ang-2 adenovirus transfection group were significantly lower than those in Ang-2 adenovirus transfection group(all P<0.05).Conclusion:Angiopoietin-2 may promote the expression of MMP-2 in endothelial cells through combining with αvβ1.
