摘要:
目的:探讨无特定病原体(SPF)级雌性昆明小鼠宫颈癌发生与阴道菌群紊乱的相关性。方法:将SPF级雌性昆明小鼠(n=48)随机平分为宫颈癌组与对照组两组。宫颈癌组在右后肢皮下注射0.2 ml宫颈癌腹水型细胞株U14悬液建立宫颈癌模型,对照组注射等剂量的生理盐水,实验观察21 d,观察阴道分泌物的菌群分布情况并进行相关性分析。结果:宫颈癌组小鼠平均第7天成瘤,成模率为100.0%。宫颈癌组建模第7、14天的每日进食量均低于对照组(均P<0.05)。对照组的阴道优势菌属为乳杆菌属,占7.2%; 宫颈癌组阴道优势菌属为巴斯德菌属和螺杆菌属,分别占26.3%和18.8%。宫颈癌组建模第7、14天的阴道菌群密集度评分均高于对照组(均P<0.05),乳杆菌半定量评分均低于对照组(均P<0.05)。宫颈癌组建模第14、21天的腹腔主动脉血液CD3+、CD4+T淋巴细胞比例均低于对照组(均P<0.05)。在建模第14天,Spearman相关性分析显示宫颈癌与阴道菌群密集度评分、乳杆菌半定量评分均存在相关性(均P<0.05)。结论:SPF级雌性昆明小鼠宫颈癌伴随有阴道菌群紊乱,可影响小鼠的免疫功能,阴道菌群紊乱介导的免疫逃逸作用可能促进了宫颈癌的发生发展。
Abstract:
Objective:To investigate the correlation between the occurrence of cervical cancer and the disorder of vaginal flora in female Kunming mice with specific pathogen free(SPF).Methods:SPF female Kunming mice(n=48)were randomly divided into cervical cancer group and control group.The cervical cancer group was injected subcutaneously with 0.2 ml of cervical cancer ascites cell line U14 suspension in the right hind limb,and the control group was injected with the same dose of normal saline.The experimental observation were 21 days,the bacterial distribution of vaginal secretions was observed and the correlation analysis was carried out.Results:The mice in the cervical cancer group developed tumors at the 7th day on average,and the model formation rate were 100.0%.The daily food intake of the cervical cancer group at the 7th and 14th day of modeling was lower than that of the control group(all P<0.05).The dominant vaginal bacteria in the control group was Lactobacillus,accounted for 7.2%; the dominant vaginal bacteria in the cervical cancer group was Pasteurella and Helicobacter,accounted for 26.3% and 18.8%,respectively.The vaginal flora density score of the cervical cancer group at the 7th and 14th day of modeling was higher than that of the control group,and the semi-quantitative score of Lactobacillus was lower than that of the control group(all P<0.05).The proportions of CD3+ and CD4+ T lymphocytes in the abdominal aortic blood at the 7th and 14th day of modeling in the cervical cancer group were lower than those in the control group(all P<0.05).At the 14th day of modeling,Spearman correlation analysis showed that cervical cancer was correlated with vaginal flora density score and Lactobacillus semi-quantitative score(all P<0.05).Conclusion:SPF female Kunming mice cervical cancer are accompanied by vaginal flora disorder,which can affect the immune function of mice.The immune escape effect mediated by vaginal flora disorder may promote the occurrence and development of cervical cancer.
