摘要:
目的:探究DEP 结构域包含5(DEPDC5)基因表达对胃癌MKN-28细胞生长及凋亡的影响。方法:流式细胞分析RNA干扰DEPDC5对胃癌细胞MNK-28凋亡的影响; 细胞划痕和Transwell检测DEPDC5对胃癌细胞侵袭迁移能力的影响。结果:与阴性对照siRNA(siRNA-NC)转染组相比,DEPDC5干扰siRNA(siRNA-DEPDC5)转染组胃癌细胞的凋亡率显著增加,迁移和侵袭能力无显著变化。结论:DEPDC5高表达与胃癌发生相关,降低DEPDC5基因表达对胃癌细胞侵袭和迁移无显著影响但可诱导胃癌细胞发生凋亡。
Abstract:
Objective:To explore the effect of dishevelled,Egl-10,and pleckstrin domain-containing protein 5(DEPDC5)gene expression on the growth and apoptosis of gastric cancer MKN-28 cells.Methods:Flow cytometry was used to analyze the effect of RNA interference DEPDC5 on the apoptosis of gastric cancer MNK-28 cells.Cell scratch and Transwell experiment was used to detect the effect of DEPDC5 on the invasion and migration ability of gastric cancer cells.Results:Compared with the negative control siRNA(siRNA-NC)group,the apoptosis rate of gastric cancer cells in the DEPDC5 interference siRNA(siRNA-DEPDC5)group was significantly increased,and there was no significant change in migration and invasion capabilities.Conclusion:The high expression of DEPDC5 is related to the occurrence of gastric cancer.Decreasing the expression of DEPDC5 gene has no significant effect on the invasion and migration of gastric cancer cells,but it can induce apoptosis of gastric cancer cells.
