摘要:
目的:探讨卷曲蛋白8(FZD8)在胃癌BGC823细胞中的表达及其对胃癌细胞迁移、侵袭的影响。方法:利用实时荧光定量PCR和Western blot检测FZD8在胃癌细胞BGC823和胃黏膜上皮细胞GES1中的表达。利用慢病毒介导的shRNA转染BGC823细胞,建立稳定的敲减FZD8基因的BGC823细胞株(shFZD8),采用Transwell实验检测胃癌细胞迁移与侵袭能力的变化。结果:BGC823细胞FZD8蛋白及mRNA表达水平均明显高于GES1细胞(均P<0.05)。shFZD8组细胞FZD8蛋白及mRNA表达水平均明显低于对照组(未敲减FZD8基因的BGC823细胞)(均P<0.05)。在BGC823细胞中下调FZD8表达能够显著抑制胃癌细胞的迁移和侵袭能力(P<0.05)。结论:FZD8在胃癌BGC823细胞中表达增加,通过shRNA抑制FZD8的表达能明显抑制胃癌BGC823细胞的迁移和侵袭。FZD8基因可能成为治疗胃癌的新靶点。
Abstract:
Objective:To investigate the expression of FZD8 in gastric cancer BGC823 cells and its effect on migration and invasion of gastric cancer cells.Methods:Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)and Western blot were used to detect the expression of FZD8 in gastric cancer cells BGC823 and normal gastric epithelial cells GES1.BGC823 cells were transfected with lentivirus mediated shRNA to establish a stable FZD8 knockdown cell line(shFZD8).Transwell assay was used to detect the changes of migration and invasion of gastric cancer cells.Results:The expression levels of FZD8 protein and mRNA in BGC823 cells were significantly higher than those in GES1 cells(all P<0.05).The expression levels of FZD8 protein and mRNA in shFZD8 group were significantly lower than those in control group(BGC823 cells without FZD8 gene knockdown)(all P<0.05).The down-regulation of FZD8 expression can significantly inhibit the migration and invasion of BGC823 cells(P<0.05).Conclusion:FZD8 expression is increased in gastric cancer BGC823 cells.Inhibition of FZD8 expression by shRNA can significantly inhibit the migration and invasion of BGC823 cells.FZD8 may become a new target for the treatment of gastric cancer.
