摘要:
目的:观察口服枸杞多糖(LBP)后肝纤维化小鼠肝组织中基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶-2抑制剂(TIMP-2)水平的变化,探究LBP干预肝纤维化的机制。方法:结扎12只小鼠胆总管造成其肝纤维化,并分为肝纤维化组和LBP组,各6只。将6只健康小鼠做为正常组。LBP组服用含有LBP 25 μg/L三蒸水200 ml,正常组和肝纤维化组仅服用三蒸水200 ml,1次/d,连续服用30 d。HE染色后比较三组小鼠肝组织结构。检测三组小鼠血清谷丙转氨酶和谷草转氨酶水平并进行比较。Western blot检测肝组织中MMP-2及TIMP-2相对表达量。结果:正常组肝脏里的每个小叶结构分界清楚,其中央为中央静脉; 肝纤维化组肝脏中有轻重程度不一、面积大小不同的组织坏死,小叶轮廓不清,只见结缔组织范围扩大,内含数量较多的小叶间胆管; LBP组肝组织坏死程度减轻,坏死面积也有所减小,结缔组织范围缩小,其内的小叶间胆管数目变少。与正常组比较,肝纤维化组小鼠血清谷丙转氨酶和谷草转氨酶水平均升高(均P<0.05)。与肝纤维化组比较,LBP组小鼠血清谷丙转氨酶和谷草转氨酶水平均下降(均P<0.05)。与正常组比较,肝纤维化组MMP-2相对表达量略微上升(P>0.05),TIMP-2相对表达量升高明显(P<0.05),MMP-2/TIMP-2下降(P<0.05)。与肝纤维化组比较,LBP组MMP-2相对表达量升高,TIMP-2相对表达量下降,MMP-2/TIMP-2上升(均P<0.05)。结论:LBP通过改变肝纤维化组织MMP-2及TIMP-2含量促使MMP-2/TIMP-2上升,从而减轻肝纤维化损伤。
Abstract:
Objective:To observe the changes of matrix metalloproteinase-2(MMP-2)and tissue inhibitor of metalloproteinase-2(TIMP-2)in liver tissue of mice with liver fibrosis after oral administration of lycium barbarum polysaccharide(LBP),and to explore the mechanism of LBP intervention in liver fibrosis.Methods:Hepatic fibrosis was induced by ligation of common bile duct in 12 mice,which were divided into hepatic fibrosis group and LBP group with 6 mice in each group.6 healthy mice were used as normal group.LBP group was treated with 200 ml triple distilled water containing 25 μg/L LBP,while normal group and liver fibrosis group were only treated with 200 ml triple distilled water,and once a day for 30 days.After HE staining,the liver tissue structure of the three groups was compared.The serum levels of alanine aminotransferase and aspartate aminotransferase were measured and compared among the three groups.The relative expressions of MMP-2 and TIMP-2 in hepatic tissue were detected by Western blot.Results:In the normal group,the structure of each lobule was clearly demarcated,and the center was the central vein; in the liver fibrosis group,there were different degrees and sizes of tissue necrosis,unclear lobule contour,only connective tissue expanded,containing a large number of interlobular bile ducts; in LBP group,the necrotic degree,necrotic area,connective tissue and the number of interlobular bile ducts were reduced.Compared with the normal group,the serum levels of alanine aminotransferase and aspartate aminotransferase were increased in the liver fibrosis group(all P<0.05).Compared with the liver fibrosis group,serum alanine aminotransferase and aspartate aminotransferase levels in LBP group were decreased(all P<0.05).Compared with the normal group,the relative expression of MMP-2 increased slightly(P>0.05),the relative expression of TIMP-2 increased significantly(P<0.05),and the ratio of MMP-2/TIMP-2 decreased(P<0.05).Compared with liver fibrosis group,the relative expression of MMP-2 was increased,relative expression of TIMP-2 was decreased,and the ratio of MMP-2/TIMP-2 was increased in LBP group(all P<0.05).Conclusion:LBP can increase the ratio of MMP-2/TIMP-2 by changing the content of MMP-2 and TIMP-2 in liver fibrosis tissue,so as to reduce the damage of liver fibrosis.
