摘要:
目的:研究烧伤后增生性瘢痕(HS)与正常皮肤(NS)组织中Toll样受体4(TLR4)及炎性细胞因子表达的水平及其相关性。方法:选取手术切除的HS组织及余留的NS组织,采用HE、Masson染色验证HS组织的特征,采用免疫组化以及实时定量PCR(RT-qPCR)检测HS组织中TLR4及炎性细胞因子的表达分布及表达水平。结果:与NS组织比较,HS组织中成纤维细胞数目、胞外基质蛋白水平、炎性分子TLR4、p65及炎性细胞因子IL-1β、IL-6、IL-8、单核细胞趋化蛋白-1(MCP-1)的mRNA水平升高(P<0.05)。相关性分析表明TLR4表达与炎性细胞因子IL-1β、IL-6、IL-8、MCP-1的表达呈正相关(P<0.05)。结论:HS组织中TLR4和炎性细胞因子水平显著高于NS,TLR4与炎性细胞因子的水平呈正相关性,表明炎症是引起HS形成的重要因素。
Abstract:
Objective:To study the expression of Toll-like receptor 4(TLR4)and proinflammatory cytokines in hypertrophic scar(HS)and normal skin(NS)tissues and their correlation.Methods:The HS and the corresponding NS tissues removed from patients who underwent surgical excision.The morphological characteristics of the HS tissue were identified by HE and Masson staining.The expression levels of TLR4 and proinflammatory cytokines were detected by immunohistochemistry and real time quantitative PCR(RT-qPCR)in HS and NS tissues.Results:Compared with NS tissue,the number of fibroblasts,the level of extracellular matrix protein,the level of TLR4 and p65,and the mRNA levels of IL-1β,IL-6,IL-8 and MCP-1 were significantly increased in HS tissue(P<0.05).The correlation analysis showed that the expression of TLR4 was positively correlated with the expression of IL-1β,IL-6,IL-8 and MCP-1(P<0.05).Conclusion:The levels of TLR4 and proinflammatory cytokines in HS tissue are significantly higher than those in NS tissue,and the levels of TLR4 and proinflammatory cytokines are positively correlated,which indicates that inflammation is an important factor in the formation of HS.
