摘要:
目的:探讨核因子-κB(NF-κB)信号通路对肺炎模型小鼠免疫功能和炎症反应的影响。方法:采用肺炎链球菌诱导肺炎小鼠模型,然后将其分为模型对照组(腹腔注射120 mg/kg生理盐水)和模型+PDTC组(腹腔注射120 mg/kg NF-κB通路抑制剂PDTC),各8只; 另取8只BALB/c小鼠作为正常对照组(腹腔注射等剂量生理盐水),均连续注射2 周后,取各组大鼠右肺上叶组织,测定湿重(W)和干重(D),并计算含水量(TLW)和干湿比(D/W)、Western blot检测各组小鼠肺组织NF-κB蛋白表达、流式细胞术检测各组小鼠外周血中T淋巴细胞亚群CD3+、CD4+、CD8+占比、ELISA试剂盒检测小鼠血清中TNF-α、IL-6和IL-10的含量。结果:①与正常对照组相比,模型对照组肺组织TLW显著增加,而D/W比值显著降低(P<0.05); 与模型对照组相比,模型+PDTC组小鼠肺组织TLW显著降低,D/W比值显著增加(P<0.05)。②与正常对照组相比,模型对照组肺组织NF-κB蛋白相对表达量显著增加(P<0.05); 与模型对照组相比,模型+PDTC组小鼠肺组织NF-κB蛋白相对表达量显著降低(P<0.05)。③与正常对照组相比,模型对照组小鼠外周血CD3+和CD4+百分比以及CD4+/CD8+比值均显著减小(P<0.05); 而与模型对照组相比,模型+PDTC组小鼠以上指标占比或比值均显著升高(P<0.05)。④与正常对照组相比,模型对照组小鼠血清促炎因子TNF-α和IL-6含量均显著升高,抗炎因子IL-10含量显著降低(P<0.05); 而与模型对照组相比,模型+PDTC组小鼠血清中TNF-α和IL-6含量均显著降低,而IL-10含量显著升高(P<0.05)。结论:肺炎的发病机制可能与NF-κB信号通路激活有关,而通过抑制该通路可显著改善肺炎小鼠肺组织病理状态,增强其免疫功能,并改善其炎症反应水平。
Abstract:
Objective:To investigate the effect of nuclear factor-kappaB(NF-κB)signaling pathway on the immune function and inflammation level of pneumonia model mice.Methods:The mice model of pneumonia were induced by Streptococcus pneumonia and divided into model control group(intraperitoneal injection of 120 mg/kg saline)and model + PDTC group(intraperitoneal injection of 120 mg/kg NF-κB pathway inhibitor PDTC),with 8 mice each.Another 8 BALB/c mice were taken as normal control group(intraperitoneal injection of equal dose of saline).After 2 weeks of continuous injection,the upper lobe tissue of the right lung of each group was taken,and the wet weight(W)and dry weight(D)were measured,and the water content(TLW)and dry-wet ratio(D/W)were calculated.Western blot was used to detect the expression of NF-κB protein in the lung tissue of each group.Flow cytometry was used to detect the proportion of T lymphocyte subsets CD3+,CD4+,and CD8+ in the peripheral blood of each group.The contents of TNF-α,IL-6 and IL-10 in the serum of the mice were detected by ELISA kit.Results:Compared with the normal control group,the lung tissue TLW of the model control group increased significantly,while the D/W ratio decreased significantly(P<0.05).Compared with the model control group,the TLW of the lung tissue of the model + PDTC group was significantly reduced,and the D/W ratio was significantly increased(P<0.05).Compared with the normal control group,the relative expression of NF-κB protein in the model control group increased significantly(P<0.05).Compared with the model control group,the relative expression of NF-κB proteinin the model + PDTC group was significantly reduced(P<0.05).Compared with the normal control group,the peripheral blood CD3+ and CD4+ percentages and the CD4+/CD8+ ratio of the model control group were significantly reduced(P<0.05).Compared with the model control group,the proportion or ratio of the above indicators in the model + PDTC group increased significantly(P<0.05).Compared with the normal control group,the serum pro-inflammatory factors TNF-α and IL-6 levels of the model control group were significantly increased,and the anti-inflammatory factor IL-10 level was significantly reduced(P<0.05).Compared with the model control group,the serum levels of TNF-α and IL-6 in the model + PDTC group were significantly reduced,while the IL-10 level was significantly increased(P<0.05).Conclusion:The pathogenesis of pneumonia may be related to the activation of NF-κB signaling pathway.Inhibiting this pathway can significantly improve the pathological state of lung tissue in mice with pneumonia,enhance their immune function,and improve the level of inflammation.
