摘要:
目的:探讨槐果碱联合奥沙利铂对裸鼠结肠癌肝转移模型的转移抑制作用。方法:利用LoVo人结肠癌细胞株建立裸鼠肝转移模型,1周后按尾静脉注入成分不同,随机分成10只槐果碱单一应用组[3.57 mg/(kg·d),SC组]、10只奥沙利铂单一应用组[1000 μg/(kg·d),Oxa组]、10只槐果碱联合奥沙利铂应用组[SC 3.57 mg/(kg·d)、Oxa 1000 μg/(kg·d),SC+Oxa组]及10只对照组(RPMI 1640 50μl,Control组)。分组制模4周后处死,观察各组药物干预后的转移情况。结果:各组裸鼠结肠癌肝转移模型成功率为100%(40/40)。与Control组相比,SC组、Oxa组及SC+Oxa组均可显著降低肝转移评分,其中SC+Oxa组更能显著抑制肝转移形成,组间比较差异有统计学意义(P<0.05)。与Control组相比,SC组、Oxa组及SC+Oxa组可显著降低肝质量、减少胆汁瘀积、血性腹水及淋巴结转移(P<0.05)。Control组局部浸润发生率和腹膜种植转移率有增高趋势,但与其他组比较差异无统计学意义(P>0.05)。结论:槐果碱联合奥沙利铂药物干预可加大抑制裸鼠结肠癌肝转移的效果。
Abstract:
Objective:To investigate the inhibitory effect of oxaliplatin in combination with sophocarpine on the hepatic metastasis of colon cancer in nude mice model.Methods:LoVo human colon cancer cell lines were used to establish the hepatic metastasis model of human colon cancer in nude mice.According to different injected components through caudal vein of tumor mice,these mice were randomly divided into sophocarpine group [3.57 mg/(kg·d),SC group],oxaliplatin group[1000 μg/(kg·d),Oxa group],sophocarpine plus Oxaliplatin [SC 3.57 mg/(kg·d),Oxa 1000 μg/(kg·d),SC+Oxa group] and control group(RPMI 1640 50 μl),10 nude mice in each group.Then four weeks later,the metastatic situation of every group were recorded after sacrificing all of mice.Results:The success rates of establishing the hepatic metastasis model of human colon cancer was 100%(40/40).Compared with the control group,the SC group,Oxa group and SC+Oxa group could significantly reduce the hepatic metastasis scores,and the SC+Oxa group can significantly inhibit the formation of hepatic metastasis,and the difference between the groups was statistically significant(P<0.05).Compared with the control group,the SC group,Oxa group and SC+Oxa group could significantly reduce liver quality,reduce cholestasis,bloody ascites and lymph node metastasis(P<0.05).The incidence of local infiltration and peritoneal implant metastasis in the control group tended to increase,but there was no significant difference compared with other groups(P>0.05).Conclusion:Sophocarpine combined with oxaliplatin drug intervention can increase the effect of inhibiting hepatic metastasis of colon cancer in nude mice.
