摘要:
目的:探讨西红花苷对糖尿病大鼠肾脏损伤的保护作用及机制。方法:雄性SD大鼠30只,按55 mg/kg一次性腹腔注射链脲佐菌素(STZ)诱导糖尿病大鼠模型。模型诱导成功后,将糖尿病大鼠随机分成糖尿病组及西红花苷组(30 mg/kg),每组15只。另取15只正常SD大鼠作为对照组。12周后,检测各组大鼠血糖、血尿素氮(BUN)、尿白蛋白/肌酐比值(ACR)、血肌酐(Scr)、丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)含量; HE染色观察大鼠肾脏病理改变,免疫组织化学染色及Western blot检测大鼠肾脏核因子κB(NF-κB)、肿瘤坏死因子-α(TNF-α)及白介素-6(IL-6)蛋白相对表达并进行比较。结果:与对照组相比,糖尿病组血糖、BUN、ACR、Scr、MDA水平明显升高,NF-κB、TNF-α及IL-6蛋白表达明显增加,SOD、CAT含量明显降低(P<0.05)。与糖尿病组相比,西红花苷组上述指标均有所改善。结论:西红花苷可对糖尿病大鼠肾脏损伤起到保护作用,其机制可能与降低血糖、抑制氧化应激和炎症反应有关。
Abstract:
Objective:To explore the protective effect and mechanism of crocin on renal injury in diabetic rats.Methods:30 male SD rats were injected intraperitoneally with streptozotocin(STZ)once at a dose of 55 mg/kg to induce diabetic rats.After the model was induced successfully,diabetic rats were randomly divided into diabetic group and crocin group(30 mg/kg),with 15 rats in each group.Another 15 normal SD rats were taken as the control group.12 weeks later,blood glucose,BUN,ACR,Scr,MDA,SOD and CAT were measured.HE staining was used to observe renal pathological changes.The relative protein expression of NF-κB,TNF-α and IL-6 were detected by immunohistochemistry staining and Western blot.Results:Compared with control group,the levels of blood glucose,BUN,ACR,Scr,MDA,and the protein expression of NF-κB,TNF-α and IL-6 were significantly increased,while the contents of SOD and CAT were decreased significantly in diabetic group(P<0.05).Compared with diabetic group,the above indexes in crocin group were improved.Conclusion:Crocin can protect the kidney of diabetic rats,and its mechanism may be related to lowering blood glucose,inhibiting oxidative stress and inflammatory response.
