摘要:
目的:检测胃癌患者的血浆中微小microRNA-100(miR-100)的表达水平,并分析miR-100水平与患者性别、年龄、肿瘤直径、有无淋巴结转移、有无远处转移、TNM分期等病理特征之间的关系。方法:采用茎环逆转录聚合酶反应(RT-PCR)方法分别检测40例胃癌患者和40例正常对照血浆中miR-100表达水平,统计分析miR-100与胃癌病理参数之间的关系。结果:正常对照组血浆中miR-100的水平为(0.58±0.18),胃癌患者血浆中 miR-100 的水平为(0.35±0.21),两组之间的差异具有统计学意义(t= 6.375,P<0.01)。胃癌患者血浆中miR-100 的水平在不同性别、不同年龄和不同肿瘤直径之间的差异无统计学意义(P>0.05); 与无淋巴转移的胃癌患者相比,有淋巴转移的患者血浆中miR-100的水平明显降低(P<0.05 ); 与无远处转移的胃癌患者相比,有远处转移的患者血浆中miR-100的水平明显降低(P<0.05 )。与Ⅰ/Ⅱ期胃癌患者相比,Ⅲ/Ⅳ期胃癌患者血浆中 miR-100的水平明显降低,比较差异具有统计学意义(P<0.05 )。结论:胃癌患者血浆中miR-100的表达水平降低,miR-100与肿瘤淋巴转移、远处转移和TNM分期有关。
Abstract:
Objective:To detect the plasma miR-100 expression level in patients with gastric cancer,and analyze the relationship between miR-100 and gender,age,tumor size,lymph node metastasis,distant metastasis and TNM stage,etc.Methods:The expression level of miR-100 in plasma of 40 gastric cancer patients and 40 normal controls was detected by stem ring reverse transcription polymerase reaction(RT-PCR),and the relationship between miR-100 and pathological parameters of gastric cancer was analyzed statistically.Results:The level of miR-100 was(0.58±0.18)in normal control group and(0.35±0.21)in gastric cancer patients.The difference between the two groups was statistically significant(t=6.375,P<0.01).There were no statistical difference in miR-100 level of gastric cancer patients between different genders,ages and tumor diameters(P>0.05).Compared with patients without lymph node metastasis,the level of miR-100 in the plasma of patients with lymph node metastasis was significantly decreased(P<0.05).Compared with patients without distant metastasis,the level of miR-100 in the plasma of patients with distant metastasis was significantly lower(P<0.05).Compared with the patients with stage Ⅰ/Ⅱ gastric cancer,the level of miR-100 in the plasma of patients with stage Ⅲ/ Ⅳ gastric cancer was decreased significantly(P<0.05).Conclusion:The expression level of miR-100 in patients with gastric cancer was decreased,and its expression level was related to tumor lymph node metastasis,distant metastasis and TNM stage.
