摘要:
目的:检测哮喘急性发作期患儿血清中微小RNA-195-5p(miR-195-5p)和脑源性神经营养因子(BDNF)的表达,分析其相关性及临床意义。方法:选择哮喘急性发作期患儿69例作为观察组,选择哮喘临床缓解期患儿69例作为对照组,选择无明显器质性疾病的儿童69例作为正常对照组,均留取血清标本,应用实时荧光定量PCR法(qPCR)检测miR-195-5p的表达,应用酶联免疫吸附实验法(ELISA)检测BDNF的表达。结果:MiR-195-5p和BDNF在三组中的表达差异有统计学意义(P<0.05),MiR-195-5p和BDNF在观察组不同病变严重程度、是否首次发病的分组中差异有统计学意义(P<0.05)。观察组中miR-195-5p与BDNF(r=-0.61,P=0.013)具有负相关性。结论: 哮喘急性发作期患儿血清中miR-195-5p低表达、BDNF高表达,参与病变的形成,二者具有负向协同作用。
Abstract:
Objective:To detect the serum expression of miR-195-5p and brain-derived neurotrophic factor(BDNF)in the serum of children with acute attack of asthma,and analyze their correlation and clinical significance.Methods:69 children with acute attack of asthma were selected as the observation group,69 children with clinical remission of asthma were selected as the control group,and 69 children without obvious organic diseases were selected as the control group.The expression level of miR-195-5p was detected by real-time fluorescent quantitative PCR.The expression level of BDNF was detected by ELISA.Results:The expression levels of miR-195-5p and BDNF were statistically different in three groups(P<0.05).In the observation group,the expression levels of miR-195-5p and BDNF were statistically different in different severity of lesions and whether the disease was the first time(P<0.05).Negative correlation was found between miR-195-5p and BDNF in the observation group(r=-0.61,P=0.013).Conclusion:Low expression of miR-195-5p and high expression of BDNF in the serum of children with acute attack of asthma are involved in the formation of pathological changes,which have a negative synergistic effect.
