摘要:
目的:研究胆管癌患者血清中微小RNA(miRNA)-244和miRNA-328的表达水平和临床意义。方法:选取肝门部胆管癌患者52例,其中胆管癌未转移患者20例,胆管癌伴转移患者32例,同期收治肝良性瘤患者43例作为对照组。采用实时定量PCR的方法,检测血清中的miRNA-244和miRNA-328的表达水平,通过ROC曲线分析miRNA-244和miRNA-328在胆管癌转移中的诊断意义。结果:胆管癌未转移组患者血清miRNA-244高于对照组(P<0.05),胆管癌转移组患者血清miRNA-244高于对照组(P<0.05); 胆管癌转移组患者血清miRNA-328低于对照组(P<0.05); 在区分胆管癌是否转移方面,miRNA-244表达水平佳界值为 4.26,此时其灵敏度和特异度分别为73.0% 和 60.1%,AUC 值为 0.7225(P=0.0161)。结论:miRNA-244在胆管癌患者中的表达水平异常升高,而miRNA-328在胆管癌患者血清中表达较低,miRNA-244对诊断和评价胆管癌是否转移有一定临床意义。
Abstract:
Objective: To study the expression and clinical significance of microRNA(miRNA)-244 and miRNA-328 in serum of patients with cholangiocarcinoma. Methods: 52 patients with hilar cholangiocarcinoma were enrolled,including 20 patients with no metastasis of cholangiocarcinoma,32 patients with metastasis of cholangiocarcinoma,and 43 patients with benign liver tumors in the same period were selected as control group. Real-time quantitative PCR was used to detect the expression levels of serum miRNA-244 and miRNA-328,and the diagnostic significance of miRNA-244 and miRNA-328 in cholangiocarcinoma metastasis was analyzed by ROC curve. Results: The level of serum miRNA-244 in the non metastatic group was higher than that in the control group(P<0.05),and that of metastatic group was also(P<0.05). The level of serum miRNA-328 in the metastatic group was lower than that in the control group(P<0.05). The optimal critical value of mirna-244 expression was 4.26,and its sensitivity and specificity were 73.0% and 60.1%,respectively. AUC was 0.7225(P=0.0161). In distinguishing whether cholangiocarcinoma had metastasized,the optimal critical value of miRNA-244 expression level was 4.26,the sensitivity and specificity were 73.0% and 60.1%,respectively,and the AUC value was 0.7225(P=0.0161). Conclusion: The expression level of miRNA-244 in patients with cholangiocarcinoma is abnormally elevated,while that of miRNA-328 is relatively low. miRNA-244 has a certain clinical significance in the diagnosis and evaluation of metastasis of cholangiocarcinoma.
