摘要:
目的:探讨微小RNA(MiR)-123调控B淋巴细胞瘤-2(Bcl-2)对宫颈癌细胞侵袭及转移能力的影响。方法:对数生长期的宫颈癌细胞Hela分为三组,MiR-123组、对照组与空白组,MiR-123组、对照组分别转染hsa-MiR-123 mimics、negative control(阴性对照),空白组转染等体积的磷酸盐缓冲液,采用CCK-8法检测细胞增殖指数,采用Transwell小室检测细胞迁移浸润指数,采用流式细胞仪检测细胞凋亡指数,采用Western blot检测Bcl-2/AKT通路蛋白表达水平。结果:细胞转染后24、36 h,MiR-123组的细胞增殖指数、侵袭及转移指数显著低于对照组与空白组(P<0.05),细胞凋亡指数显著高于对照组与空白组(P<0.05),对照组与空白组对比差异无统计学意义(P>0.05)。细胞转染后36 h,MiR-123组的Bcl-2、AKT蛋白相对表达水平显著低于对照组与空白组(P<0.05),对照组与空白组对比差异无统计学意义(P>0.05)。结论:过表达MiR-123能抑制Bcl-2/AKT通路的激活,从而抑制宫颈癌细胞增殖、侵袭及转移,促进细胞凋亡,从而发挥抑癌作用。
Abstract:
Objective:To investigate the effect of miR-123 regulating B-cell lymphoma-2(Bcl-2)on the invasion and metastasis of cervical cancer. Methods:The cervical carcinoma hela cells in logarithmic growth phase were divided into MiR-123 group,control group and blank group. MiR-123 group and control group were transfected with hsa-MiR-123 mimics and negative control respectively,and the blank group was transfected with phosphate buffer solution. The cell proliferation index was detected by CCK-8 method. The cell migration and infiltration index was detected by the Transwell chamber. The apoptosis index was detected by flow cytometry. The Bcl-2/AKT protein expression level was detected by Western blot. Results:24 hours and 36 hours after cell transfection,the cell proliferation index,migration and infiltration index of the MiR-123 group were significantly lower than those of the control group and the blank group(P<0.05),and the apoptosis index was significantly higher than that of the control group and the blank group(P<0.05),and there were no significant difference between the control group and the blank group(P>0.05). 36 hours after cell transfection,the relative expression levels of Bcl-2 and AKT proteins in the MiR-123 group were significantly lower than those in the control group and the blank group(P<0.05),and there were no significant difference between the control group and the blank group(P>0.05). Conclusion:Overexpression of MiR-123 can inhibit the activation of Bcl-2/AKT pathway,so as to inhibite the proliferation,invasion and metastasis of cervical cancer cells,promote cell apoptosis,and thus exert tumor suppressing effect.
