摘要:
目的:观察丁苯酞(NBP)对阿尔茨海默病(AD)模型大鼠海马CA1区脑源性神经营养因子(BDNF)和碱性成纤维生长因子(bFGF)表达的影响。方法:72只雄性SD大鼠随机分为假手术组(Sham组)、AD模型组(AD组)、丁苯酞治疗组(NBP组)。采用向双侧海马区注射β-淀粉样蛋白1-42(Aβ1-42)制作AD模型。采用水迷宫实验检测AD大鼠的学习记忆能力,免疫组化法、Western blot法检测海马CA1区BDNF和bFGF蛋白表达水平。结果:与Sham组比较,AD组大鼠出现明显的学习记忆障碍,各时间点海马CA1区BDNF和bFGF 蛋白表达增多(P<0.01); 与AD组比较,NBP组大鼠学习记忆障碍明显改善,各时间点海马CA1区BDNF和bFGF蛋白表达明显增多(P<0.01),以4周时表达最明显。结论:丁苯酞能改善AD大鼠的学习记忆能力,其机制可能与丁苯酞上调BDNF和bFGF蛋白表达有关。
Abstract:
Objetive:To observe the effect of dl-3-n-butylphthalidle(NBP)on the expression of brain-derived neurotrophic factor(BDNF)and basic fibroblast growth factor(bFGF)in CA1 region of hippocampus of Alzheimer disease(AD)rats. Methods: Totally 72 SD rats were randomly divided into sham-operation group(Sham group),AD model group(AD group)and NBP treatment group(NBP group). The AD rat model was prepared by injecting beta-amyloid protein 1-42 into bilateral hippocampus. The ability of learning and memory of AD rats were tested by water maze test,and the expression of BDNF and bFGF in hippocampal CA1 region were detected by immunohistochemistry and Western blot. Results: Compared with the Sham group,the AD group showed significant learning and memory impairment,and the expression of BDNF and bFGF in CA1 region of hippocampus were significantly increased at each time point(P<0.01). Compared with the AD group,the learning and memory impairment of NBP group was significantly improved,and the expression of BDNF and bFGF in CA1 region of hippocampus were significantly increased at each time point(P<0.01),especially at week 4. Conclusion: NBP can improve the learning and memory ability of AD rats,which may be related to the up regulation of BDNF and bFGF protein expression.
