目的:探讨外源性重组人白细胞介素11(rhIL-11)促进肿瘤转移,而白藜芦醇抑制肺腺癌A549细胞转移灶形成,为进一步研究提供理论依据。方法:经裸鼠尾静脉注入肺腺癌A549细胞,建立肿瘤转移模型,同时使用不同剂量rhIL-11单药或联合白藜芦醇进行干预,比较各组出现转移灶的裸鼠数量和各组转移灶数量的差异。结果:对照组有1只裸鼠出现转移灶,转移率为16.7%(1/6),低剂量rhIL-11组有3只裸鼠出现转移灶,转移率为50.0%(3/6),高剂量rhIL-11组有4只裸鼠出现转移灶,转移率为66.7%(4/6)。 rhIL-11[750 μg/(kg?d)]+白藜芦醇组和rhIL-11 [500 μg/(kg?d)]+白藜芦醇组出现转移灶的裸鼠数少于rhIL-11组[750 μg/(kg?d)]和rhIL-11组[500 μg/(kg?d)](3/12对7/12),比较差异无统计学意义(χ2=2.743,P=0.098)。且转移灶个数也少于rhIL-11组(5个对13个)。结论:rhIL-11可能促进肿瘤转移灶的形成,而白藜芦醇可能有抑制rhIL-11促肿瘤转移灶形成作用。
Abstract:
Objective:To investigate whether exogenous recombinant human interleukin 11(rhIL-11)can promote tumor metastasis,and whether resveratrol can inhibit the formation of metastases of lung adenocarcinoma A549 cells,and provide theoretical basis for further research. Methods:Lung adenocarcinoma A549 cells were injected into the tail vein of nude mice to establish a tumor metastasis model,and different doses of rhIL-11 alone or in combination with resveratrol were used for intervention,and the number of nude mice with metastatic lesions and the number of metastatic lesions in each group were compared. Results:The metastasis rate was 16.7%(1/6)in the control group,50.0%(3/6)in the low-dose rhIL-11 group and 66.7%(4/6)in the high-dose rhIL-11 group. The number of nude mice with metastatic lesions in rhIL-11 + resveratrol groups [rhIL-11 750 μg/(kg·d)+ resveratrol group and rhIL-11 500 μg/(kg·d)+ resveratrol group] was less than that in rhIL-11 groups [rhIL-11 750 μg/(kg·d)group and rhIL-11 500 μg/(kg·d)](3/12 vs.7/12),but the difference was not statistically significant(χ2=2.743,P=0.098). The number of metastatic lesions in rhIL-11 + resveratrol groups was also less than that in rhIL-11 groups(5 vs.13). Conclusion:rhIL-11 may promote the formation of tumor metastasis,while may be inhibited by resveratrol.
