摘要:
目的:探讨过表达miRNA-224骨髓间充质干细胞(BMSCs)对卵巢颗粒细胞的作用及其机制。方法:构建miRNA-224慢病毒载体,转染BMSCs,qRT-PCR法验证转染效率。将转染后的BMSCs与卵巢颗粒细胞共培养后分为六组:对照组、BMSCs组、miRNA-224-BMSCs组、顺铂组、BMSCs+顺铂组和miRNA-224-BMSCs+顺铂组。四甲基偶氮唑蓝(MTT)法检测细胞存活率,丙二醛(MDA)和活性氧(ROS)试剂盒检测氧化应激水平。结果:qRT-PCR法结果表明,转染后BMSCs miRNA-224的表达显著上调,差异有统计学意义(P<0.05); 与对照组相比,顺铂组细胞存活率显著降低,差异有统计学意义(P<0.05),而与顺铂组相比,BMSCs+顺铂组和miRNA-224-BMSCs+顺铂组细胞存活率显著升高,且相较于BMSCs+顺铂组,miRNA-224-BMSCs+顺铂组细胞存活率进一步升高,差异有统计学意义(P<0.05); 与对照组相比,顺铂组细胞MDA和ROS水平升高,差异有统计学意义(P<0.05),而与顺铂组相比,BMSCs+顺铂组和miRNA-224-BMSCs+顺铂组细胞MDA和ROS水平显著降低,且相较于BMSCs+顺铂组,miRNA-224-BMSCs+顺铂组细胞MDA和ROS水平进一步降低,差异有统计学意义(P<0.05)。结论:过表达miRNA-224的BMSCs可明显降低顺铂诱导的卵巢颗粒细胞凋亡,其机制与氧化应激有关。
Abstract:
Objective:To explore the effect and mechanism of bone marrow mesenchymal stem cells(BMSCs)overexpressing miRNA-224 on ovarian granulosa cells.Methods:The miRNA-224 lentiviral vector was constructed and transfected into BMSCs.The transfection efficiency was verified by qRT-PCR.The transfected BMSCs were co-cultured with ovarian granulosa cells and divided into six groups:control group,BMSCs group,miRNA-224-BMSCs group,cisplatin group,BMSCs+cisplatin group and miRNA-224-BMSCs+cisplatin group.Cell survival rate was measured by MTT assay,and oxidative stress level was measured by malondialdehyde(MDA)and reactive oxygen species(ROS)kits.Results:The results of qRT-PCR showed that the expression of miRNA-224 was significantly up-regulated after transfection(P<0.05).Compared with the control group,the cell survival rate of the cisplatin group was significantly lower(P<0.05).Compared with the cisplatin group,the cell survival rate of the BMSCs+cisplatin group and the miRNA-224-BMSCs+cisplatin group was significantly higher(P<0.05).Moreover,the cell survival rate of the miRNA-224-BMSCs+cisplatin group was further higher than that of the BMSCs + cisplatin group(P<0.05).Compared with the control group,the levels of MDA and ROS in the cisplatin group was significantly higher(P<0.05).Compared with the cisplatin group,the levels of MDA and ROS in the BMSCs+cisplatin group and the miRNA-224-BMSCs+cisplatin group was significantly lower(P<0.05).Moreover,the levels of MDA and ROS in the miRNA-224-BMSCs+cisplatin group was further lower than that of the BMSCs+cisplatin group(P<0.05).Conclusion:BMSCs overexpressing miRNA-224 may obviously reduce ovarian granulosa cell apoptosis induced by cisplatin,and its mechanism is related to oxidative stress.
