摘 要 目的:研究甲状腺全切除术对甲状腺癌患者血清血管内皮生长因子(VEGF)、可溶性Fas受体(sFas)、可溶性Fas受体配体(sFasL) mRNA表达的影响。方法:纳入拟行甲状腺全切除术治疗的甲状腺癌患者40例作为研究对象。分别于术前1 d以及术后1 d采集所有受试者的静脉血10 ml,采用反转录聚合酶链反应(RTPCR)法检测VEGF、sFas、sFasL mRNA表达水平,以酶联免疫吸附法检测血清白细胞介素6(IL6)、白细胞介素8(IL8)、肿瘤坏死因子α(TNFα)水平,并作相关性分析。结果:手术后患者血液样本中VEGF、sFas、sFasL mRNA表达水平分别为(0.54±0.02)、(0.38±0.02)、(0.37±0.02),显著低于手术前的(0.76±0.01)、(0.60±0.04)、(0.59±0.04)(均P<0.05)。手术后患者血清白细胞介素6(IL6)、白细胞介素8(IL8)、肿瘤坏死因子α(TNFα)水平分别为(173.29±16.93)pg/ml、(0.57±0.01)ng/L、(1.39±0.03)ng/L,显著低于手术前的(321.28±24.82)pg/ml、(2.71±0.02)ng/L、(3.41±0.32)ng/L(均P<0.05)。经Pearson相关性分析可得:甲状腺癌患者血液中VEGF、sFas、sFasL mRNA水平与血清IL6、IL8、TNFα水平均呈正相关关系(均P<0.05)。结论:甲状腺全切除术可显著下调甲状腺癌患者的VEGF、sFas、sFasL mRNA表达水平,同时下调IL6、IL8、TNFα水平。
Abstract:
Abstract Objective:To study the effect of total thyroidectomy on mRNA expression levels of vascular endothelial growth factor (VEGF),soluble Fas receptor (sFas) and soluble Fas receptor ligand (sFasL) in serum of patients with thyroid cancer. Methods:40 patients with thyroid cancer planned to undergo total thyroidectomy were enrolled as study subjects. 10 ml of venous blood was collected from all subjects one day before surgery and one day after surgery,and the mRNA expression levels of VEGF,sFas,and sFasL were measured by RTPCR. The levels of IL6,IL8,and TNFα were detected by enzymelinked immunosorbent assay,and correlation analysis was performed. Results:The mRNA expression levels of VEGF,sFas,and sFasL in the blood samples of patients after surgery were (0.54±0.02),(0.38±0.02),and (0.37±0.02),respectively,which were significantly lower than those before surgery (0.76±0.01),(0.60±0.04),and (0.59±0.04) (all P<0.05). The levels of serum IL6,IL8,and TNFα were (173.29±16.93) pg/ml,(0.57±0.01) ng/L,and (1.39±0.03) ng/L after surgery,respectively,which were significantly lower than those before surgery (321.28±24.82) pg/ml,(2.71±0.02) ng/L,and (3.41±0.32) ng/L (all P<0.05). Pearson correlation analysis showed that the mRNA expression levels of VEGF,sFas and sFasL in the blood of patients with thyroid cancer were positively correlated with the levels of serum IL6,IL8 and TNFα (all P<0.05). Conclusion:Total thyroidectomy can significantly downregulate the mRNA expression levels of VEGF,sFas and sFasL,as well as the levels of IL6,IL8 and TNFα in patients with thyroid cancer.
