摘要:
摘 要 目的:探讨五味子乙素在大鼠离体心脏缺血/再灌注损伤模型中的保护作用及对心肌糖酵解反应的调节作用。方法:采用Langendorff离体心脏灌流方法,建立心肌缺血/再灌注体外模型,通过血流变仪考察心肌血流动力学指标;采用酶标仪检测心肌组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPx)活性及丙二醛(MDA)含量,检测心肌组织中半胱氨酸天冬氨酸蛋白酶3(Caspase3)活性,ATP含量及乳酸含量。结果:五味子乙素对缺血/再灌注后大鼠离体心脏血流动力学具有显著影响,可显著升高左室发展压(LVDP)、左室内压上升/ 下降速率(±dp/dt)和心率(HR);同时明显降低心肌组织中氧化应激反应的发生,减少MDA 含量,升高SOD及GSHPx抗氧化酶活性,此外可明显抑制心肌组织中Caspase3,同时升高心肌组织中ATP及乳酸含量,但以上作用与糖酵解抑制联合使用后显著降低。结论:五味子乙素可提高心肌糖酵解水平,抑制心肌缺血再灌注导致的氧化应激反应,减少心肌细胞线粒体损伤的发生,恢复心肌能量供应并改善缺血再灌注心脏血流动力学。
Abstract:
Abstract Objective:To investigate the protective effect of schisandrin B on isolated rat heart model of ischemiareperfusion injury and its regulation on myocardial glycolysis. Methods:An in vitro model of myocardial ischemia/reperfusion was established by Langendorff method. The hemodynamic parameters of myocardium were measured by hemorheology instrument. The activities of superoxide dismutase(SOD),glutathione peroxidase(GSHPx) and the content of malondialdehyde(MDA) in myocardium were measured by enzyme labeling instrument. The activity of cysteine aspartate protease(caspase3),ATP content and lactic acid content in myocardium were measured. Results:Schisandrin B had a significant effect on the hemodynamics of isolated rat heart after ischemia/reperfusion,which could significantly increase left ventricular development pressure(LVDP),left ventricular pressure rise/fall rate(±dp/dt) and heart rate(HR). at the same time,it could significantly reduce the occurrence of oxidative stress response in myocardial tissue,reduce MDA content,increase SOD and GSHPx antioxidant enzyme activity. in addition,it could significantly inhibit caspase3,increase the contents of ATP and lactate in cardiac tissue,but the above effects were significantly reduced in combination with glycolysis inhibition. Conclusion:Schisandrin B can improve the level of myocardial glycolysis,inhibit the oxidative stress response caused by myocardial ischemiareperfusion,reduce the occurrence of myocardial mitochondrial damage,restore the energy supply of myocardium and improve the hemodynamics of myocardial ischemiareperfusion. 
