摘 要 目的:探讨黄连素治疗对老年大鼠心肌梗死后的保护作用及其机制。方法:40只老年大鼠,随机分为心肌梗死盐水对照组和心肌梗死黄连素治疗组。将所有老年大鼠的左冠状动脉前降支中上1/3进行永久性结扎,术后第2天,腹腔注射盐水和黄连素,连续注射28 d,并记录死亡情况和体重变化。在第7天,取心脏组织检测炎症水平,观察指标包括大鼠整合素αM型(CD11b),白介素6(IL6),趋化因子( Ccl2),肿瘤坏死因子α(TNFα)。在第28天,用超声评估大鼠心脏功能,并对心脏组织进行HE染色和Masson染色。结果:与盐水对照组相比,黄连素治疗组的病死率明显降低,体重无明显差异。黄连素治疗后,梗死后的老年大鼠的心脏功能明显恢复,左室射血分数(LVEF)显著升高(P<0.01),左室短轴缩短率(LVFS)明显降低(P<0.001)。而且,形态学分析结果提示长期的黄连素治疗可以显著减少心室重塑,HE染色结果说明黄连素治疗可以保护心肌细胞结构的完整性。同样的,Masson染色提示黄连素治疗可以减少心肌组织纤维化程度。另外,在黄连素治疗后7 d,损伤心脏的炎症水平较盐水对照组明显降低,表现为CD11b阳性单核细胞数量减少,促炎因子IL6和TNFα基因水平显著增加,炎症趋化因子Ccl2水平也增加。最后,黄连素组的磷酸肌醇3激酶(PI3K)水平增高,苏氨酸激酶(AKT)水平下降。结论:黄连素治疗可以促进老年大鼠心梗后的心脏功能恢复,其机制可能与激活了PI3K/AKT通路发挥了抗炎作用有关,这为黄连素的临床应用提供了实验基础。
Abstract:
Abstract Objective:To investigate the protective effect of berberine on myocardial infarction in aged rats and its mechanism.Methods:40 aged rats were randomly divided into two groups:myocardial infarction saline control group and myocardial infarction berberine treatment group.The left anterior descending coronary artery of all aged rats was permanently ligated.On the second day after operation,saline and berberine were injected intraperitoneally for 28 consecutive days.Mortality and weight changes were recorded.On the 7th day,the levels of inflammation were measured in heart tissues,including CD11b,IL6,Ccl2 and TNFalpha.On the 28th day,the cardiac function was assessed by ultrasonography,and the cardiac tissue was stained with HE and Masson staining.Results:Compared with the saline control group,the mortality rate of berberine treatment group was significantly lower,but there was no significant difference in body weight.After berberine treatment,the cardiac function of the aged rats recovered significantly,the LVEF increased significantly(P<0.01),and the LVFS decreased significantly(P<0.001).Moreover,morphological analysis showed that berberine treatment could significantly reduce ventricular remodeling in the long term.HE staining results showed that berberine treatment could protect the integrity of myocardial cell structure.Similarly,Masson staining suggests that berberine treatment can reduce the degree of myocardial fibrosis.In addition,at 7 days after berberine treatment,the inflammation level of injured heart was significantly lower than that of saline control group,which showed that the number of CD11bpositive monocytes decreased,the levels of proinflammatory factors IL6 and TNFalpha genes increased significantly,and the level of inflammatory chemokine Ccl2 also increased.Finally,PI3K level increased and AKT level decreased in Berberine group.Conclusion:Berberine treatment〖LM〗 can promote the recovery of cardiac function after myocardial infarction in aged rats,and its mechanism may be related to the activation of PI3K/AKT pathway to play on antiinflammatory role,which lays an experimental foundation for the clinical application of berberine.
