摘要:
摘 要 目的:研究妊娠期糖尿病(GDM)对成年子代小鼠糖脂代谢的影响及其作用机制。方法:选取8周龄健康雌性与雄性小鼠按照1∶1的比例合笼喂养,通过阴栓检测法和阴道精子检测法明确小鼠是否受孕。将受孕的小鼠20只按照随机抽签法等分成观察组及对照组,观察组以120 mg/kg的剂量予以腹腔注射链脲霉素建立GDM小鼠模型,对照组则采用同等剂量的柠檬酸柠檬酸钠缓冲液干预。比较两组子代小鼠出生体重、空腹血糖、血浆瘦素水平,分娩后20周葡萄糖耐量试验(GTT)与胰岛素耐量试验(ITT)结果,血脂指标水平,肝脏相关基因表达情况。结果:观察组子代小鼠的出生体重、空腹血糖均高于对照组,而血浆瘦素水平低于对照组(均P<005)。分娩后20周观察组GTT实验30、60、90 min时成年子代小鼠的血糖水平高于对照组(均P<005)。分娩后20周观察组成年子代小鼠总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDLC)水平均高于对照组,而高密度脂蛋白胆固醇(HDLC)水平低于对照组(均P<005)。分娩后20周观察组成年子代小鼠ITT试验30、60、90 min时的血糖水平高于对照组(均P<005)。观察组成年子代小鼠过氧化物酶体增殖物激活受体(PPARα)、肉毒碱棕榈酰基转移酶1α(CPT1α)、羟基酰基辅酶A脱氢酶(Ehhadh)相对表达量均高于对照组,而长链酯酰辅酶A合成酶3(ACSL3)相对比表达量低于对照组(均P<005)。结论:GDM会引起子代成年后糖脂代谢紊乱,其主要作用机制可能与肝脏相关基因表达异常有关。
Abstract:
Abstract Objective: To investigate the effect of gestational diabetes mellitus(GDM) on glucose and lipid metabolism in adult offspring mice and its mechanism Methods: Eightweekold healthy female and male mice were selected and fed in cages in the ratio of 1∶1 Twenty of pregnant mice were divided into observation group and control group by random lottery method The observation group was intraperitoneally injected with streptozotocin at the dose of 120 mg/kg,while the control group was intraperitoneally injected with sodium citrate buffer at the same dose The birth weight,fasting blood glucose and plasma leptin levels of offspring mice in two groups were compared At 20 weeks after delivery,the results of GTT test and ITT test,the levels of blood lipid indicators,and the expression of liver related genes were compared Results: The birth weight and fasting blood glucose of adult offspring mice in the observation group were higher than those in the control group,while the plasma leptin level was lower (all P<005) At 20 weeks after delivery,blood glucose levels of adult offspring mice in the observation group were higher than those in the control group in the GTT and ITT test of 30 min,60 min and 90 min (all P<005);the TC,TG and LDLC levels of adult offspring mice in the observation group were higher than those in the control group,while HDLC level was lower (all P<005);the relative expression levels of PPARα,CPT1α and Ehhadh in adult offspring mice were higher in the observation group than those in the control group,while ACSL3 was lower (all P<005)Conclusion: GDM can induce glucose and lipid metabolism disorder in adult offspring mice,and the main mechanism may be related to abnormal expression of liver related genes
