摘 要 目的:探讨卡托普利对高血压大鼠炎性因子的影响及是否可以逆转心室肥厚的不良结局,并对炎性因子与心室肥厚的相关性进行研究。方法:自发性高血压大鼠(SHR)随机分为SHR对照组(SHR组,n=7),卡托普利组(Cap组,n=7);7只SD大鼠为常规对照组(CON组),Cap组给予卡托普利 25 mg/kg;SHR组和CON组给予等量的生理盐水;1次/d,持续12周;测干预后各组大鼠第0周、4周、8周、12周的血压变化;测各组大鼠超敏C反应蛋白(hsCRP)、肿瘤坏死因子α(TNFα)、内皮素1(ET1)、一氧化氮(NO)等相关炎性因子及心脏重量指数变化情况。结果:①SHR大鼠血压显著高于对照组(P<0.001);SHR大鼠给药后,其血压随着时间的延长显著下降,至第8周后,血压趋于稳定,给予卡托普利的SHR大鼠收缩压显著下降;②SHR大鼠给予卡托普利后,SHR组大鼠血清TNFα、ET1、 HsCRP相关炎性因子及心脏重量指数显著降低,NO水平显著升高,趋于正常对照CON组水平。③心脏重量指数与TNFα、ET1及hsCRP有显著的正相关(P<0.001);与NO有显著的负相关(P<0.001)。结论:卡托普利可以改善SHR大鼠的炎性反应,并可通过降低炎性反应抑制心肌肥厚的发生;同时利用相关炎性反应可以预测大鼠心室肥厚的程度。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗Objective :To investigate the effect of captopril on inflammatory factors in hypertensive rats and whether it can reverse the adverse outcome of ventricular hypertrophy;and to study the correlation between inflammatory factors and ventricular hypertrophy.Methods:SHR rats were randomly assigned to SHR control group (SHR group,n=7),captopril group (Cap group,n=7);7 SD rats were used as regular control group (CON group),Cap group was given Cato Pu Li 25 mg/kg;SHR group and CON group were given the same amount of normal saline;once a day for 12 weeks;blood pressure changes at 0,4,8 and 12 weeks after the intervention;The changes of hsCRP,TNFα,ET1,NO and other related inflammatory factors and heart weight index were observed in each group.Results:①The blood pressure of SHR rats was significantly higher than that of the control group (P<0.001).After SHR rats were administered,their blood pressure decreased significantly with time.After the eighth week,the blood pressure became stable,systolic blood pressure was significantly decreased in SHR rats given captopril.②After administration of captopril in SHR rats,the serum TNFα,ET1,HsCRPrelated inflammatory factors and heart weight index of SHR group were significantly decreased.The level of NO increased significantly and tended to be normal in the CON group.③There was a significant positive correlation between cardiac weight index and TNFα,ET1 and HsCRP (P< 0.001);there was a significant negative correlation with NO (P<0.001).Conclusion:Captopril can improve the inflammatory response in SHR rats and inhibit the occurrence of cardiac hypertrophy by reducing the inflammatory response.At the same time,the inflammatory response is used to predict the degree of ventricular hypertrophy in rats.
