摘要:
摘 要 目的:探讨醒脑静对大鼠脑出血病灶周围星形胶质细胞表达水通道蛋白4(AQP4)的影响。方法:建立胶原酶VII定位注射诱发大鼠尾壳核脑出血模型,随机分成醒脑静治疗组和对照组。建模后给予一次药物治疗。醒脑静治疗组大鼠给予腹腔注射醒脑静注射液2 ml/kg,对照组大鼠给予腹腔注射等容量生理盐水。治疗72 h后对比两组大鼠生存率、行为学改变,处死取材测定脑组织含水量变化,观察脑实质组织学及星形胶质细胞超微结构的改变;逆转录PCR检测AQP4 mRNA的表达;蛋白免疫印迹检测AQP4蛋白的表达。含血清培养大鼠星形胶质细胞,随机分成5 ml/L、10 ml/L、20 ml/L醒脑静组和对照组,培养基中分别加入5 ml/L、10 ml/L、20 ml/L的醒脑静和等容量的生理盐水,逆转录PCR检测AQP4 mRNA的表达;蛋白免疫印迹检测AQP4蛋白的表达。结果:醒脑静治疗组大鼠生存率、行为学评分均较对照组明显改善;电镜下显示醒脑静治疗可减轻模型大鼠病灶周围星形胶质细胞水肿,上调脑组织表达AQP4蛋白与mRNA的表达,细胞实验显示,随着培养基中加入醒脑静浓度增加,星形胶质细胞的AQP4蛋白与mRNA的表达增加。结论:经醒脑静治疗后的脑出血大鼠无论在生存率还是在肢体对称平衡方面均得到改善,其机制可能是醒脑静可直接刺激星形细胞上调AQP4蛋白及mRNA的表达,从而缓解脑出血后血管源性脑水肿的进展,这为未来醒脑静用于治疗脑出血后脑水肿治疗提供新的理论支持。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗Objective:To investigate the effect of AQP4 on the expression of astrocytes in peripheral astrocytes of rats afterintracerebral hemorrhage.Methods:The model of intracerebral hemorrhage in rat caudate nucleus was established by Collagenase VII location injection,which were randomly divided into the treatment group and the control group.The rats in the treatment group were given intraperitoneal injection of 2ml/kg,and the control group gave intraperitoneal injection and other capacity saline.After 48h treatment,the survival rate and behavioral changes of two groups of rats were compared,and the changes of water content in brain tissue were measured,and the changes of brain parenchyma and astrocytes ultrastructure were observed.The expression of AQP4mRNA was detected by RTPCR,and the expression of AQP4 protein was detected by Western blot.Serum cultured rat astrocytes were randomly divided into 5ml/l,10ml/l,20ml/l and control group,respectively,5ml/l,10ml/l and 20ml/l were added to the brain static and equal capacity of normal saline,and RTPCR was detected to express the AQP4 mRNA;The expression of AQP4 protein was detected by Western blot.Results:The survival rate and behavioral score of the group were significantly improved compared with those in the control group.Under electron microscope,it can alleviate the cell edema of astrocytes around the lesion of the model rats,increase the expression of AQP4 protein and mRNA in the brain tissue,and the cell experiment shows that the concentration of awake brain increases with the addition of the culture medium.The expression of AQP4 protein and mRNA in astrocytes increased.Conclusion:The cerebral hemorrhage rats after the treatment of cerebral ischemia have been improved in both survival rate and limb symmetry balance,the mechanism may be that the brain can directly stimulate the expression of AQP4 protein and mRNA,and 〖LM〗thus alleviate the development of vascular brain edema after intracerebral hemorrhage,This provides a new theoretical support for the treatment of cerebral edema after intracerebral hemorrhage in the future.
