摘 要 〖HT5K〗目的:探讨SCNN1B基因与人胃癌细胞上皮间质转化的相关性。方法:构建SCNN1B过表达载体,稳定转染人胃癌SGC7901细胞。于裸鼠右前肢皮下接种细胞,制备胃癌裸鼠移植瘤模型。3周后处死裸鼠,观察移植瘤生长情况。分别采用Realtime PCR、Western blot、免疫组化检测移植瘤组织中SCNN1B、Ecadherin,Ncadherin及Vimentin表达情况。结果:SCNN1B过表达抑制胃癌移植瘤增长,提高Ecadherin表达水平,降低Ncadherin和Vimentin表达水平。结论:过表达SCNN1B能够抑制胃癌细胞的生长及上皮间质转化。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗Objective:To investigate the correlation between SCNN1B gene and epithelialmesenchymal transformation of human gastric cancer cells. Methods:SCNN1B overexpression vectors were constructed and the vectors were stably transfected into human gastric cancer SGC7901 cells. The nude mice were injected subcutaneously with cells to establish a transplanted tumor model of human gastric carcinoma. After three weeks,nude mice were executed and growth situation of cancer xenografts in the nude mice was observed. The expression of SCNN1B,Ecadherin,Ncadherin and Vimentin in transplanted tumors were detected by Realtime PCR,Western blot and immunohistochemistry. Results:Overexpression of SCNN1B inhibited the growth of gastric cancer xenografts as well increased the expression of Ecadherin and decreased the expression of Ncadherin and Vimentin. Conclusion:SCNN1B overexpression can inhibit the gastric cancer cell growth and epithelial mesenchymal transition.
