摘 要 〖HT5K〗目的:探究脉冲电磁场(PEMF)改善db/db小鼠肝脏脂肪变性的潜在机制,以期为PEMF用于改善非酒精性脂肪性肝病(NAFLD)提供实验依据。方法:通过C57BL/KsJ db/db小鼠构建NAFLD模型,随机分为PEMF组和NAFLD组,对照组由8只C57BL/KsJ小鼠组成。PEMF组每天施加外源性PEMF(20 Gs,1 h)刺激,持续12周,其他组不做任何处理。实验结束后检测空腹血糖、血清胰岛素,计算HOMAIR;检测肝脏甘油三酯、丙二醛、还原性谷胱甘肽及氧化型谷胱甘肽、谷胱甘肽过氧化物酶;检测SREBP1c蛋白表达量。结果: PEMF组胰岛素抵抗水平降低约25%;丙二醛及氧化型谷胱甘肽水平显著下降,而还原性谷胱甘肽与谷胱甘肽过氧化酶水平明显上升;SREBP1c的蛋白表达量较NAFLD组减少约35%左右;脂滴数量和大小均显著降低;肝脏TG含量降低至(4.49±0.86) mmol/gprot。结论:PEMF通过调控SREBP1c及氧化应激改善db/db小鼠肝脏脂肪变性。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗〖JP2〗Objective:The potential mechanism for pulsed electromagnetic fields (PEMF) improves the liver steatosis in db/db mice was investigated. Methods:The 8 C57BL/KsJ mice were as control group and 16 C57BL/KsJ db/db mice were randomly divided into PEMF group and NAFLD group. The PEMF group was given exposure of 15.38 Hz frequency and 20 Gs field intensity 1 h/d for 12 weeks. Then the blood glucose and insulin levels were detected to calculate the HOMAIR. The levels of triglyceride (TG),malonaldehyde (MDA),glutathione (GSH) and oxidized glutathione(GSSG) and glutathione peroxidase(GSHPx) were determinated. The expression of SREBP1c protein was determinated. Results:Compared with NAFLD group,the HOMAIR of PEMF group decreased 25% (P<0.05). Besides,liver steatosis was also ameliorated. Further explorations revealed that PEMF could reduce the expression of SREBP1c and increase the expression of GSHPx and consequently suppress oxidative stress. Conclusion:PEMF attenuates oxidative stress and suppresses the expression of SREBP1c to ameliorate liver steatosis in db/db mice. 〖JP〗
