摘 要 目的:探讨结直肠癌缺失基因(DCC)和丝裂原活化蛋白激酶激酶4(MKK4)在人结直肠癌组织中的表达及临床意义。方法:应用逆转录聚合酶链反应(RT-PCR)法、蛋白质印记(Western-blot)法、免疫组织化学法,对60例结直肠癌组织及对应癌旁组织中的DCC、MKK4基因表达水平进行检测,并结合患者临床病理因素分析DCC、MKK4表达与临床病理特征的关系。结果:结直肠癌组织中DCCmRNA、MKK4mRNA基因表达水平均低于癌旁组织(P<0.05);结直肠癌组织中DCC、MKK4蛋白表达水平均低于癌旁组织(P<0.05);结直肠癌组织中DCC、MKK4蛋白表达的阳性率低于癌旁组织(P<0.05);结合患者临床病理参数发现,DCC、MKK4蛋白表达水平均与肿瘤分化程度、淋巴转移、远处转移有显著相关性(P<0.05)。结论:DCC、MKK4是结直肠癌的抑制基因,在结直肠癌的发生、发展及转移等过程中发挥一定作用。
Abstract:
Abstract Objective: To investigate the expression and clinical significance of delected in colorectal carcinoma (DCC) and Mitogen-activated pro tein kinase kinase 4 (MKK4) in human colorectal cancer. Methods: The expression levels of DCC and MKK4 genes in 60 colorectal cancer tissues and corresponding adjacent tissues were detected by RT-PCR,Western-blot and immunohistochemistry. The relationship between DCC,MKK4 and clinicopathological characteristics was analyzed in combination with clinical and pathological factors. Results: The results of RT-PCR showed that the expression of DCC,MKK4 in colorectal cancer tissues were lower than those in adjacent tissues (P<0.05). Western-blot showed that the expression levels of DCC,MKK4 in colorectal cancer tissues were significantly lower than those in adjacent tissues (P<0.05). Immunohistochemistry showed that the expression levels of DCC,MKK4 in colorectal cancer tissues were lower than those in adjacent tissues (P<0.05). According to the clinical case factors, the expression levels of DCC/MKK4 were significantly correlated with the degree of differentiation, lymphatic metastasis and distant metastasis (P<0.05). Conclusion: DCC,MKK4 the suppressor genes of colorectal cancer and play a role in the occurrence, development and metastasis of colorectal cancer.
