摘 要 目的:研究藏红花素对视网膜缺血再灌注损伤的保护作用。方法:将SD大鼠随机分为四组:正常对照组、缺血再灌注模型组,藏红花素给药高剂量和低剂量组。造模前3 d藏红花素给药组动物给予藏红花素腹腔注射,1次/d。通过提高眼内压复制视网膜缺血再灌注模型。再灌注后24 h处死动物,迅速摘除眼球并固定视网膜。免疫组织化学染色法观察胶质细胞酸性蛋白(GFAP)在视网膜的表达量,使用电镜观察视网膜细胞结构。结果:模型组大鼠视网膜GFAP表达量显著增加,该组大鼠视网膜超微结构为Müller细胞突起增多,电子密度增加以及中间丝蛋白成束排列。藏红花素给药高剂量组\[50 mg/(kg·d)\]GFAP表达量较模型组显著降低。结论:藏红花素能通过阻止Müller活化有效保护视网膜免受缺血再灌注损伤。
Abstract:
Abstract Objective:To investigate the crocin anti-apoptosis for retinal cells in rat retina ischemia-reperfusion (I/R) model. To test whether the crocin given counteracts the effects of I/R injury in the rat retina.Methods:Four experimental groups of Sprague-Dawley rats were used,normal control,ischemia-reperfusion,crocin -treated high and low dose groups. Animals in treated group were injected intraperitoneally with crocin once a day for 3 days before ischemia. Retinal I/R injury was induced by raising the intraocular pressure (110 mmHg) for 1 hour. 24 hours after ischemia,the animals were killed and the retinas were quickly removed,fixed. Expression levels of glial fibrillary acidic protein (GFAP)were investigated by immunohistochemistry. Electron microscopy was used to examine the structural alterations in retinal cells. Results:In the model group ,An increase in Müller cell glial fibrillary acidic protein (GFAP) levels was observed at 1 day after ischemia. And cellular pathologies of Muller cells including swollen processes,increase of electron density and bundles of intermediate filament. The expression of GFAP in high dose group \[50 mg/(kg·d)\] of crocin administration was significantly lower than that of ischemia-reperfusion group. Conclusion:Crocin can effectively protect the retina from ischemia-reperfusion injury by preventing the activation of Müller cells. 
